摘 要:目的探讨人参皂苷对骨髓间充质干细胞(bone marrow mesenchymal stem cell,BMSC)分泌干细胞因子的影响。方法通过密度梯度离心法从健康志愿者抗凝骨髓中分离、培养BMSC,传代3代后,采用流式细胞术鉴定其细胞表型;MTT法检测不同浓度人参皂苷(50、100、150、200、300、400 mg/ml)作用不同时间(24、48、72、96 h)对BMSC增殖活性的影响;ELISA检测200 mg/ml人参皂甙作用不同时间(24、48、72 h)对BMSC分泌干细胞因子SCF-1和干细胞衍生因子SDF-1α水平的影响;RT-PCR法检测200 mg/ml人参皂甙作用24 h对BMSCSCF-1和SDF-1αmRNA转录水平的影响。结果第3代BMSC的CD44、CD29和CD105表达阳性,而CD34、CD45和HLA-DR表达阴性,BMSC的纯度为90%;BMSC与不同浓度的人参皂苷共培养不同时间,细胞的增殖活性均明显增强(P<0.05),200 mg/ml人参皂苷作用96 h细胞的增殖活性最高(P<0.05),但无明显的时间和剂量依赖性;人参皂苷对BMSC SCF-1和SDF-1α的分泌及mRNA的转录具有明显的促进作用。结论人参皂苷能增强BMSC分泌干细胞因子,有望应用于辅助干细胞治疗。Objective To investigate the effect of ginsenoside on secretion of stem cell factors by bone marrow mesenchy- mal stem cells (BMSCs). Methods BMSCs were isolated from the bone marrow of healthy volunteers by density gradient centrifugation, then cultured and subcultured for three passages, and identified for phenotype by flow cytometry. The proliferation activities of BMSCs stimulated with ginsenoside at various concentrations (50, 100, 150, 200, 300 and 400 mg/ml) for various (24, 48, 72 and 96) hours were determined by MTT method. The method of RT-PCR was used to detect the secretion of stem cell cytokines. The secretion levels of stem cell factor SCF-1 and stem cells-derived factor SDF-lot by BMSCs after treatment with 200 mg/ ml ginsenoside for 24, 48 and 72 h were determined by ELISA. The transcription levels of SCF-1 and SDF-IoL in BMSCs after treatment with 200 mg/ml for 24 h were determined by RT- PCR. Results BMSCs of passage 3 were positive for CD44, CD29 and CD105, while negative for CD34, CD45 and HLA-DR, of which the purity was 90%. The proliferation activities of BMSCs increased significantly after co-culture with ginsenoside at various concentrations for various hours (P 〈 O. 05), of which that after co-culture with 200 mg/ml gin- senoside for 96 h was the highest (P 〈 O. 05). However, the proliferation activity was time- and dose-independent. Gin- senosdie promoted the secretions and mRNA transcriptions of SCF-1 and SDF-β in BMSCs. Conclusion Ginsenoside enhanced the secretion of stem cell factors by BMSCs, which might be used for accessory stem cell therapy.
