摘  要:目的:探讨益精方对腺嘌呤法大鼠肾虚型不育症动物模型的调整作用。方法:SPF级健康成年雄性Wistar大鼠,1.5月龄,体重(180±10)g,共60只,正常饲养1周,随机分为5组。每组12只,分别为空白组、模型组、大剂量组、中剂量组、小剂量组,将腺嘌呤按500 mg/ml浓度配制,按与腺嘌呤1∶10比例加入阿拉伯胶作为助溶剂,1 ml/(100 g·d)剂量灌胃10 d造模结束。治疗48 d后观察其肾虚证相关表现、精子浓度和活力、睾酮等激素水平、性腺及附属性腺重量等指标的变化,并进行组间比较分析。结果:各组动物的一般状态及精子密度和活力、睾酮水平、性腺和附属性腺重量、动物体重等各项相关指标及表现均有明显的调整作用(P<0.05)。按空白组、模型组、益精方大剂量组、中剂量组和小剂量组的顺序:精子浓度为(87.85±28.44)×106/ml、(7.11±2.15)×106/ml、(35.98±14.04)×106/ml、(32.65±11.80)×106/ml、(33.51±13.26)×106/ml;精子活率为(52.79±16.43)%、(31.14±3.07)%、(45.88±16.97)%、(51.56±13.35)%、(49.53±10.16)%;睾酮为(194.07±40.29)nmol/L、(61.27±13.70)nmol/L、(121.87±24.35)nmol/L、(127.44±19.38)nmol/L、(127.81±20.28)nmol/L;黄体生成素为(7.02±0.27)IU/L、(6.12±0.89)IU/L、(7.06±0.87)IU/L、(7.16±0.94)IU/L、(6.97±0.78)IU/L;睾丸的重量为(3.78±0.18)g、(2.87±0.26)g、(3.24±0.06)g、(3.46±0.07)g、(3.40±0.09)g;附睾重量为(1.12±0.12)g、(0.83±0.23)g、(1.12±0.10)g、(1.13±0.11)g、(1.11±0.11)g;前列腺重量为(176.75±427.09)mg、(131.67±39.45)mg、(178.70±37.97)mg、(180.11±37.39)mg、(179.00±35.42)mg;体重为(188.50±7.12)g、(189.92±6.67)g、(187.42±5.47)g、(189.17±6.19)g、(188.75±6.12)g。睾丸组织病理显示:模型组可见明显损伤,益精方各剂量组均有改善。结论:益精方对腺嘌呤法大鼠肾虚型不育症动物模型具有明显的治疗作用。Objective: To investigate the regulatory effect of Yijing Fang (YJF) on adenine-induced infertility in rats with kid-ney deficiency. Methods: Sixty healthy Wistar male rats, aged 1.5 mo and weighing ( 180 ± 10) g, were normally fed for a week, and then divided into five groups of equal number (blank control, infertile model, high-dose YJF, mid-dose YJF, and low-dose YJF) according to the body weight of the rats. The models were made by intragastric administration of 500 mg/ml adenine in gum arabic solu- tion in the ratio of 1:10 at the dose of 1 ml per 100 g body weight per day for 10 days. YJF was given at 3.38 g, 1.69 g and 0.85 g per 100 g body weight per day to the rats in the high-, mid- and low-dose groups, respectively. After 48 days of treatment, we observed kidney deficiency-related changes in sperm concentration and motility, the levels of testosterone (T) and other hormones and the vol- umes of the testis, epididymis, seminal vesicle and prostate, and compared the indexes among different groups. Results: YJF exhibi- ted a significant regulatory effect on sperm concentration and motility, the T level and the indexes of the gonad and other accessory glands in the model rats (P 〈 0.05 ). After 48 days of treatment, sperm concentrations were ( 87.85 ± 28.44 ), (7.11 ± 2.15 ), (35.98 2 14.04), (32.65 2 11.80) and (33.51 2 13.26) x 106/ml in the blank control, infertile model, high-dose YJF, mid-dose YJF, and low-dose YJF groups, respectively; sperm motilities were (52.79 2 16.43), (31.14 2 3.07), (45.88 ± 16.97), (51.56 ±13.35) and (49.53 ±10.16)%; the Tlevels were (194.07 ±40.29), (61.27±13.70), (121.87±24.35), (127.44±19.38) and (127.81±20.28) nmol/L; the luteinizing hormone (LH) levels were (7. 017 ±0.269), (6. 117 ±0.894), (7. 060 ± 0.871), (7.156±0.937) and (6.967 ±0.778) IU/L; the testis volumes were (3.775 ±0.183), (2.865 ±0.258), (3.236 ± 0.058), (3.457 �
