摘 要:目的:考察温度及pH对甘草酸溶液中内毒素团聚状态的影响。方法:采用30 kDa Millipore膜过滤不同温度及pH条件下甘草酸溶液,通过动态浊度法测定超滤液中内毒素活性浓度,HPLC检测超滤前后有效成分的变化,利用内毒素纳米粒径仪测定不同温度及pH条件下内毒素团聚状态的粒径分布。结果:温度60℃,pH 4时,内毒素团聚状态的粒径分布和去除率均最大,甘草酸溶液经30 kDa Millipore膜超滤前后有效成分含量>98%。结论:随温度的升高,甘草酸溶液中内毒素团聚状态的粒径分布变大,而随pH的增大则变小。Objective: To investigate effects of temperature and pH on agglomeration state of endotoxin in glycyrrhizic acid solution.Method: Under different temperature and pH conditions,glycyrrhizic acid solution was filtered with 30 kDa Millipore membrane,the concentration of Endotoxin in ultrafiltrate was determined by dynamic turbidity method,contents change of active components was examined by HPLC,particle size of agglomeration state of endotoxin was determined with nanometer particle size instrument under different temperature and pH conditions.Result: When temperature was 60 ℃ and pH was 4,particle size distribution and removal of endotoxin agglomeration state were optimal.The contents of active components in glycyrrhizic acid solution was above 98% after filtering by 30 kDa Millipore membrane.Conclusion: With increasing of temperature,particle size distribution and removal of endotoxin agglomeration state increased,but when pH increased from 4 to 10,particle size and removal decreased.
