摘  要:目的:采用HPLC-ELSD测定预知子中常春藤皂苷元3-O-β-D-吡喃葡萄糖基-(1→3)-α-L-吡喃鼠李糖基-(1→2)-α-L-吡喃阿拉伯糖苷(皂苷X,saponins X),常春藤皂苷元3-O-β-D-吡喃木糖基-(1→2)-α-L-吡喃阿拉伯糖苷(皂苷B,saponins B),常春藤皂苷元3-O-α-L-吡喃鼠李糖基-(1→2)-α-L-吡喃阿拉伯糖苷(皂苷P D,saponins P D),常春藤皂苷元3-O-α-L-吡喃阿拉伯糖苷(皂苷A,saponins A)的含量。方法:采用安捷伦1100手动高效液相色谱仪,色谱柱为Cosmosil-C18ODS色谱柱(4.6mm×250 mm,5μm),ELSD检测器,流动相乙腈-0.2%甲酸水(45∶55),体积流量1.0 mL·min-1;蒸发光散射检测器检测参数:漂移管温度98℃,氮气流量2.8 L·min-1。结果:皂苷X,皂苷B,皂苷P D,皂苷A进样量在2~10μg线性关系良好(R2分别为0.999 0,0.999 0,0.999 1,0.999 1)。平均回收率(n=6)分别为96.8%,101.9%,99.3%,100.5%;RSD分别为0.9%,1.2%,0.7%,1.3%。结论:该方法简单,分离效果好,结果准确,可用于预知子药材中皂苷X,皂苷B,皂苷P D,皂苷A的含量测定。Objective: To establish a method for the determination of the content of 3-O-β-Dglucopyranosyl(1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl hederagenin(saponins X),3-Oβ-D-xylopyranosyl-(1→2)-α-L-arabinopyranosyl hederagenin(saponins B),3-O-α-L-rhamnopyranosyl-(1→ 2)-α-L-arabinopyranosyl hederagenin(saponins P D),3-O-α-L-arabinopyranoside hederagenin(saponins A) in Akebiae Fructus using high-performance liquid chromatography with evaporative light scattering detector(HPLCELSD).Method: The separation of A.Fructus was performed on an Agilent 1100 manual high performance liquid chromatograph,Cosmosil-C 18 ODSchromatography column(4.6 mm × 250 mm,5 μm).The mobile phase was acetonitrile-water and the flow rate was 1.0 mL·min-1.The detector was ELSD.The temperature of drift tube was 98 ℃.And the nebulizer nitrogen flow rate was 2.8 L·min-1.Result: The linear range of saponin X,saponin B,saponins P D,saponin A was 2-10 μg(R2was 0.999 0,0.999 0,0.999 1,0.999 1,respectively),and the average recovery(n = 6) was 96.8%,101.9%,99.3%,100.5%,respectively;and RSD was 0.9%,1.2%,0.7%,1.3%,respectively.Conclusion: The method is simple,reliable and accurate.It can be used to determine saponins X,saponins B,saponins P D,saponins A content and control the quality in Akebiae Fructus.
