摘  要:目的:建立高效液相色谱法比较桂枝不同商品规格化学成分差别,探讨中药材商品规格与药材化学成分的相关性。方法:采用Waters-Symmetry-RP-C18(4.6 mm×250 mm,5μm)色谱柱,乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长275 nm,柱温30℃。结果:香豆素、肉桂醇、肉桂酸、桂皮醛线性范围分别为0.001 32~0.132μg(r=0.999 5),0.001 08~0.108μg(r=0.999 6),0.002 57~0.257μg(r=0.999 6),0.141 4~1.448μg(r=0.999 7);平均加样回收率分别为97.8%(RSD 1.5%),97.2%(RSD 1.1%),100.6%(RSD 1.3%),99.6%(RSD 1.7%)。不同规格桂枝药材中香豆素、肉桂醇、肉桂酸与桂皮醛含量差别较大。结论:方法准确,重复性好,简便易行,可有效地评价桂枝药材的质量,为桂枝药材的商品流通和开发研究提供可靠的依据。Objective: To compare different chemical composition of Cinnamomi Ramulus specifications by HPLC method and discuss the relationship between specifications and chemical compositions.Method: WatersSymmetry-RP-C 18 column(4.6 mm × 250 mm,5 μm) was used.The mobile phase consisted of solvent A(acetonitrile) and solvent B(0.1% aqueous phosphoric acid) by gradient elution.The flow rate was 1.0 mL· min-1.The wavelength was set at 275 nm.The column temperature was maintained at 30 ℃.Result: The calibration curves of coumarin,cinnamyl alcohol,cinnamic acid and cinnamaldehyde were in good linearity over the ranges of 0.001 32-0.132 μg(r = 0.999 5),0.001 08-0.108 μg(r = 0.999 6),0.002 57-0.257 μg(r = 0.999 6),0.141 4-1.448 μg(r = 0.999 7),and the average recoveries were 97.8%,97.2%,100.6%,99.6% with RSD of 1.5%,1.1%,1.3%,1.7% respectively.The content of coumarin,cinnamyl alcohol,cinnamic acid and cinnamaldehyde in Cinnamomi Ramulus of different specifiations was different.Conclusion: This method is simple,sensitive and accurate,which will supply evidence for the quality control and research of Cinnamomi Ramulus.
