摘 要:目的:观察柴胡-白芍不同比例配伍应用对实验性惊厥小鼠的影响。方法:将雄性昆明种小鼠随机分为7组,即模型组(A),阳性药对照组(B),柴胡-白芍2∶1组(C),柴胡-白芍1∶1组(D),柴胡组(E),白芍组(F),柴胡-白芍1∶2组(G)。模型组给予等量生理盐水,阳性药对照组用量为苯妥英钠0.025 g·kg-1或地西泮0.004 g·kg-1,柴胡-白芍不同比例配伍组用量为10.4 g·kg-1,各组小鼠连续ig给药7 d,末次给药60 min后造模,观察各用药组对小鼠最大电惊厥(MES)模型、戊四氮模型、士的宁模型及匹鲁卡品模型的阵挛潜伏期、强直潜伏期、死亡潜伏期及死亡数的影响。结果:与A组相比较,在MES模型中,B,F组可以显著降低惊厥发生率(P<0.05);在戊四氮模型中,B,C,D组均能显著延长实验小鼠的阵挛潜伏期、强直潜伏期(P<0.05),B,C组能显著延长实验小鼠的死亡潜伏期(P<0.05);在士的宁模型中,B组能够显著延长实验小鼠的阵挛潜伏期、死亡潜伏期(P<0.05),其余各组对实验小鼠的阵挛潜伏期、死亡潜伏期,无显著影响;在匹罗卡品模型中,B,C,D,E,G组均能显著延长实验小鼠的死亡潜伏期(P<0.05)。结论:柴胡-白芍配伍应用具有良好的抗惊厥的作用,其抗惊厥作用优于单用柴胡或白芍,其中以柴胡-白芍2∶1配伍比例的抗惊厥作用最好。Objective: To observe the anticonvulsant effect of Bupleuri Radix(BR) and Paeoniae Radix Alba(PA) with different compatibility proportion in mice.Method: The Kunming mice were randomly divided into seven groups: the model group(A),the positive control group(B),BR-PA2: 1 group(C),BR-PA1: 1group(D),BR group(E),PA group(F),RB-PA1: 2 group(G).The positive control group received the administration ofphenytoin 0.025 g·kg-1or diazepam 0.004 g·kg-1,the dosage of BR and PA with different compatibility proportion was 10.4 g·kg-1,and the model group received the same amount of saline.After the continuous lavage administration for 7 days,we tested the anticonvulsant effect of BR and PA with different compatibility proportion in mouse models of maximal electroskock(MES),pentylenetetrazole(PTZ),strychnine and pilocarpine-induced convulsions 60 min after the last administration,the latency of clonic convulsion,tonic convulsion and death were observed.Result: In MES model,compared with group A,group B and F could significantly decrease the incidence of seizures(P 0.05);in PTZ model,group B,C,D could significantly prolong the latency of clonic,convulsions and tonic convulsions(P 0.05),group B,C could obviously prolong the latency of death;in strychnine model,all groups except group B(P 0.05) had no effect on the latency of clonic and death;in pilocarpine model,compared with group A,all groups could prolong the latency of death(P 0.05).Conclusion: Bupluri Radix and Paeoniae Radix Alba with application compatibility display commendable anticonvulsant effect,which is better than Bupluri Radix or Paeoniae Radix Alba alone,anticonvulsant effect of BR and PA group with ratio of 2∶ 1 is best.
