摘 要:目的探讨18β-甘草次酸对2型糖尿病大鼠血糖、血脂及胰岛素敏感性的影响。方法采用高脂高糖饲料诱导结合一次性小剂量注射链脲佐菌素的方法建立2型糖尿病大鼠模型。将大鼠随机分为正常对照组(CON)、正常给药组(CGA)、糖尿病组(DM)、糖尿病+甘草次酸治疗组(DGA)、糖尿病+格列苯脲阳性对照组(DGL),采用灌胃方式分别给予溶剂(5%DMSO)、18β-甘草次酸[100 mg/(kg·d)]和格列苯脲[600μg/(kg·d)]。45 d后,检测实验大鼠的空腹血糖(FBG)、血脂(TG、TC、LDL-C和HDL-C)以及血清胰岛素。结果18β-甘草次酸显著降低了糖尿病大鼠的FBG、TG、TC、LDL-C和胰岛素抵抗指数(HOMA-IR),并升高了其抗动脉粥样硬化指数(AAI)。结论 18β-甘草次酸能有效降低2型糖尿病大鼠的血糖和血脂,改善胰岛素抵抗。[ Objective ] To explore the effect of 18β-glycyrrhetinic acid on fasting blood glucose (FBG), lipids and sensitivity of insulin in type 2 diabetic rats. [Methods ] Type 2 diabetes model was replicated by high-fat- high-sugar diet combined with lower dose STZ injection. All rats were randomly divided into five groups: control group(CON), control 18β-glycyrrhetinic acid treatment group (CGA), diabetes group (DM), DM+18β-glycyrrhetinic acid treatment group (DGA) and DM+glibenclamide treatment group (DGL). They were orally administered with 5% DMSO, 18β-glycyrrhetinie acid[100mg/(kg. d)] and glibenclamide [600μg/(kg. d)] respectively. After 45 days of con- tinuous medication, detect fasting blood glueose(FBG), lipids(TG, TC, LDL-C, HDL-C) and insulin level in serum of all rats. [Results] 18β-glycyrrhetinic acid treatment significantly decreased the levels of FBG, TG, TC, LDL-C and homeostasis model assessment of insulin resistance (HOMA-IR) in diabetic rats, and also increased anti- atheroselerosis index(AAI). [ Conclusion] 18β-glycyrrhetinic acid has hypoglycemic and hypolipidemic effects,and it can also ameliorate insulin resistance.
