摘  要:目的 探讨糖尿病大鼠治疗前后逼尿肌和腰骶背根神经节(DRG)中神经生长因子(NGF)的表达及意义.方法 将30只SD大鼠随机分为5组,正常对照组6只,糖尿病组24只;建立糖尿病大鼠模型,将其随机分为4组:糖尿病组6只、单用NGF组6只、单用胰岛素组6只、联合应用NGF和胰岛素组6只,治疗4周后,应用免疫组织化学方法检测各组大鼠逼尿肌和腰骶背根神经节中NGF的表达.结果 对照组血糖明显低于糖尿病组[(30.90±2.33) mmol/L比(4.30±0.18) mmol/L(t =30.33,P<0.01)];糖尿病大鼠经NGF和胰岛素治疗后膀胱和腰骶背根神经节中NGF的含量较非治疗组明显增加(0.1492±0.0046比0.1066±0.0055,0.1667±0.0065比0.1174±0.0052,P<0.05).结论 NGF在糖尿病大鼠膀胱病变的发病过程中具有重要作用,给予外源性NGF可能有助于糖尿病大鼠膀胱病变病变减轻和功能改善.Objective To investigate the expression of nerve growth factor (NGF) in lumbar dorsal root ganglia (DRG) and bladder of streptozotocin-induced diabetic rats before and after treatment.Methods Thirty male Sprague-Dawley (SD) rats were divided into five groups randomly,including 6 in control group,24 in diabetic cystopathy (DC) group.The rat DC models were made throught injection of STZ.The rats in DC group were divided into four subgroups randomly (n =6 each),including DC group,NGF group,RI group,and NRG + RI group.The diabetic rats were treated with NGF and insulin for 4 weeks.NGF was detected in the bladder and DRG by using immunohistochemical staining.Results The blood sugar level in DC group was significantly higher than that in the control group [(30.90 ± 2.33) vs.(4.30 ±0.18) mmol/L (t =30.33,P < 0.01)].The NGF levels in both bladder and DRG of diabetic rats treated with NGF and insulin were significantly increased (0.1492 ± 0.0046 vs.0.1066 ± 0.0055,P<0.05; 0.1667 ±0.0065 vs.0.1174 ±0.0050,P<0.05).Conclusion The aberrant expression of NGF in both bladder and DRG of diabetic rats may play an important role,and using exogenous NGF may help to alleviate the pathological changes and improve the function of rats with DC.
