摘 要:目的:采用血清药理学方法探讨皂角刺含药血清诱导肠癌细胞凋亡的作用机制。方法:制备皂角刺药液,50只健康SD大鼠分为皂角刺高中低剂量组(每1 mL分别含有生药2 000,1 000,500 mg),环磷酰胺阳性对照组(50 mg·kg-1)和生理盐水空白对照组5个组,按20 mL·kg-1大鼠体质量给药,以制备含药血清。取含药血清培养液在体外作用于SW-480细胞(密度为1×106/mL),分别在24,48,72 h后,采用MTT法检测含药血清对SW-480增殖的影响;加入药物血清培养液处理细胞24 h后,采用流式细胞仪检测SW-480凋亡率,RT-PCR检测各组含药血清对细胞B细胞淋巴瘤/白血病2(Bcl-2)/Bel-2相关x蛋白(Bax)基因表达的影响。结果:与空白对照组比较,皂角刺含药血清培养液作用细胞的吸光度(A)在20,40,60 h均明显低于空白组(0.370±0.005)%,(0.624±008)%,(1.078±0.038)%(P<0.01);皂角刺高、中、低剂量含药血清培养液诱导SW-480凋亡依次为(37.386±0.163)%,(35.834±0.092 3)%,(27.572±1.193)%显著高于空白对照(5.036±0.378)%(P<0.01);皂角刺含药血清培养液对SW-480细胞的Bcl-2表达(0.258±0.037,0.442±0.024和0.652±0.072)明显低于空白对照组(0.936±0.047)(P<0.01),随灌胃给药的药物剂量增加Bcl-2表达而增加,而Bax表达(1.946±0.017,1.810±0.023和1.810±0.023)明显高于空白对照组(1.260±0.029)(P<0.01),Bax表达反而减少。结论:皂角刺含药血清对能抑制SW-480生长和诱导其凋亡,Bcl-2 mRNA基因表达减少与Bax mRNA基因表达增多可能是皂角刺诱导肠癌细胞凋亡的机制之一。Objective: To study the effect of serum containing Gleditsiae Spina on inducing apoptosis and B cell lymphoma/leukemia-2 (Bcl-2) and Bcl-2 associated x protein (Bax) mRNA expression of human colorectal cancer SW-480 cells. Method: Liquid extracted from Gleditsiae Spina was prepared, the serum containing Gleditsiae Spina was prepared by feeding healthy 50 SD rats at high, medium and low dose of Gleditsiae Spina (2 000, 1 000, 500 mg·L^-1), normal saline and cyclophosphamide (CTX) group (50 mg ·kg^-1 ) according to the rat body weight (20 mL ·kg^-1 ). The impact of serum containing Gleditsiae Spina on SW-480 with a cell density of 1 x 106/mL proliferation was estimated by methyl thiazolyl tetrazolium (MTT) at 24, 48, 72 h after than that of the blank group (5. 036 ±0. 378)% (P 〈0.01) , the expression of mRNA of Bax (0. 258 ±0. 037, 0. 442 ±0. 024, 0. 652 ±0. 072) was significantly lower than that of the blank group (0. 936± 0. 047 ) (P 〈 0.01), whereas expression of Bcl-2 (1.946 ±0.017, 1.810±0.023, 1.724±0.028) was higher than that of the blank group ( 1. 260 ± 0. 029) (P 〈 0.01 ). Conclusion : The serum containing Gleditsiae Spina can inhibit proliferation and induce apoptosis of human colorectal cancer SW-480 cells. The decreased expression of Bcl-2 gene and the increased expression of Bax gene may be the mechanism of apoptosis of SW-480.
