摘  要:目的:探讨蓝萼香茶菜总二萜预处理对兔心肌缺血/再灌注损伤的保护作用。方法:健康日本大耳白兔24只,分4组,A:假手术组;B:缺血/再灌注(I/R)组;C:复方丹参滴丸(DS)对照组;D:蓝萼香茶菜总二萜(TD)组。术前7 d A,B组灌胃蒸馏水5 mL·kg-1·d-1,C组灌胃DS 37.8 mg·kg-1·d-1,D组灌胃TD 4.76 mg·kg-1·d-1。末次给药1 h后,结扎左冠状动脉前降支(LAD)制作兔在体心肌I/R模型。光镜和电镜观察各组心肌细胞结构变化。原位末端标记法(TUNEL)检测心肌细胞凋亡,免疫组化法测定心肌组织凋亡相关蛋白B细胞淋巴瘤/白血病-2(Bcl-2)和Bcl-2相关x蛋白(Bax)的表达。结果:C组和D组心肌细胞凋亡指数(AI)分别为10.93%和9.87%,与B组(18.40%)相比,AI均明显下降(P<0.05),但C组和D组无明显差异。免疫组化分析Bcl-2的吸光度(A),C组为41.03,D组为52.09,均明显高于B组(29.47),有显著性差异(P<0.05)。而与B组相比,C组和D组Bax的A明显下降(P<0.05),且C组和D组Bcl-2和Bax无明显差异。与假手术组相比,B组光镜和电镜下结构损伤程度明显高于C组和D组。结论:TD能减轻兔心肌缺血/再灌注损伤时心肌细胞凋亡,提示该药对心肌有保护作用。Objective: To study the effects of total diterpenoids from Rabdosia japonica var. glaucocalyx (TD) preconditioning on myocardial ischemia/reperfusion injury (MI/RI). Method: Twenty-four rabbits were randomly divided into four groups, sham group (group A) , I/R group (group B) , Danshen dripping pills (DS) group (group C) and TD group (group D). Group A and B were administered distilled water (5 mL·kg^-1·d^-1) intragastrically daily for 7 days prior to the operation. Rabbits in group C, D underwent the same surgical procedure as those in group B, but received respectively oral DS (37.8 mg ·kg^-1·d^-1 ) and TD (4.76 mg ·kg^-1·d^-1 ) , 7 days before the operationl Uhrastructure was observed by light microscopy and electron microseopy. TUNEL staining detected myocardial cell apoptosis index. Immunohistoehemisty determined expression of apoptosis-related protein B cell lymphoma/leukemia-2 (Bcl-2) and Bcl-2 associated x protein (Bax). Result: Apoptosis index of myocardial cells in group C and D is respectively 10.93% and 9.87% , compared with group B ( 18.40% ) the difference was obvious (P 〈 0.05 ). Immunohistopathological studies showed that A of Bcl-2 in group C, D was 41.03, 52.09, compared with group B (29.47) (P 〈 0.05). Conclusion: TD may reduce MI/RI in rabbit model and have protective effects on mvocardiuml
