摘  要:Tilling(Targeting induced local lesions in genomes)技术是近年来兴起的利用化学诱变或者物理诱变方法产生一系列的点突变,经过PCR快速筛选突变体,进而验证基因功能的方法。近年来,Tilling技术被用于多种生物的研究中。对小麦这种基因组庞大、遗传背景复杂的作物来说,Tilling技术在分析基因功能、挖掘重要基因资源和改良作物品质方面比传统的转基因方法有更大的优势。然而,Tilling技术在小麦中的应用还存在筛选工作量大等问题。本文介绍了Tilling技术的操作流程、问题改进及在小麦基因功能研究和品质改良方面的研究现状。Tilling (Targeting induced local lesions in genomes) technology is a new strategy in recent years that uses chemical mutagenesis method or physical mutagenesis method to create libraries of mutagenized individuals, and then uses PCR (polymerase chain reaction) to amplify and screen an allelic series of induced single base substitutions in target gene. Through the above experiments, Tilling technology can be used in gene function analysis at last. In recent years, Tilling technology is used in the study of a variety of plants, animals, as well as organisms. As far as wheat is concerned, which with a huge genomes and a complex background, Tilling technology has more advantages than the traditional transgenic method in the gene function analysis,identification of important gene and crop quality improvement. However, there are still large screening workload and other problems in Tilling technology application in wheat. In this review, the technical processes, research improvement of Tilling are introduced. Moreover, the application of Tilling in wheat gene function analysis and improvement are reviewed.
