摘 要:目的:考察苦参碱脂质体的稳定性及体外释放度。方法:用硫酸铵梯度法制备苦参碱脂质体,考察其粒径、形态和包封率;以渗漏率为指标考察苦参碱脂质体在4℃和室温放置9个月内的稳定性;同时采用桨碟法考察苦参碱脂质体的体外释放情况。结果:制备的苦参碱脂质体的平均粒径为220 nm,包封率为63.37%,4℃放置9个月的渗漏率为(10.7±0.1)%。脂质体的体外释放规律符合Higuchi方程。结论:苦参碱脂质体应在冷藏条件下贮存,其在体外具有良好的缓释作用。OBJECTIVE: To study the stability and release rate in vitro of Matrine liposome. METHODS: The ammonium sul- fate gradient method was applied to produce the liposome, and the diameter, morphology and entrapment efficiency were studied. Using leakage rate as index, the stability of Matrine liposome at 4 ℃ and room temperature was investigated within 9 months, and the release behavior of the drug was studied by paddle over disk method. RESULTS: The average particle size of Matrine liposome was 220 nm and entrapment efficiency was 63.37%; the leakage rate of drug was (10.7 _+ 0.1)% at 4 ℃ within 9 months. The re- lease rule in vitro was up to the Higuchi equation. CONCLUSIONS: Matrine liposome should be kept in cold storage and has sus- tained-release effect.
