摘  要:目的建立雏血宁中虎杖苷、大黄素-8-O-β-D-葡萄糖苷、大黄素、大黄素甲醚4种化学成分的含量测定的HPLC方法。方法采用Kromasil C18(4.6mm×250mm,5p,m)色谱柱;0.1%的磷酸溶液和乙腈进行梯度洗脱;柱温30℃;流速1mL·min^-1;检测波长223nm。结果在HPLC法中,虎杖苷、大黄素-8-O-β-D-葡萄糖苷、大黄素、大黄素甲醚的线性范围分别是:0.009~0.072mg·mL^-1(r=0.9991)、0.048~0.386mg·mL^-1(r=1.0000)、0.027~0.214mg·mL^-1(r=1.0000)、0.024~0.188mg·mL^-1(,=1.0000);平均回收率分别为98.8%、97.1%、95.5%、96.8%,RSD分别为0.6%、0.4%、0.3%、0.4%。结论所建立的方法专属性强,重复性好,可用于维血宁的质量控制。Objective To establish an HPLC method for the simultaneous determination of polydatin, emodin-8-O-β-D- glycosidase, emodin and physcion in Weixuening. Methods The chromatograhic separation was performed on Kromasil C18 column (4.6 mm × 250 mm, 5 μm). The column temperature was 30 ℃. A linear elution of 0.1% phosphoric acid aqueous solu- tion-acetonitrile was adopted at the flow rate of 1 mL . min^-1. The UV detection wavelength was 223 nm. Results Polydatin, emodin-8-O-β-D-glycosidase, emodin and physcion had good linearity at 0.009 -- 0.072 mg .mL ^- 1(r = 0.999 1), 0.048 -- 0.386 mg . mL^ -1 (r= 1.000 0), 0.027 -- 0.214 mg .mL^-1 (r = 1.000 0), and 0.024-0.188 mg . mL^- 1 (r = 1.000 0), respectively. The average recoveries and RSD were 98.8% (0.6%), 97.1% (0.4%), 95.5% (0.3%), and 96.8% (0.4%), respectively. Conclusion This method is simple, accurate and reproduceable, which can be used for the quality control of Weixuening.
