摘　　要：目的建立黄芪桂枝五物汤中的黄芪甲苷的含量测定方法。方法采用HPLC-ELSD法测定黄芪桂枝五物汤中黄芪甲苷的含量,色谱条件为：Welchrom-C18色谱柱（250mm×4.6mm,5μm）,以乙腈-水（32∶68）为流动相,流速为1mL·min-1,柱温25℃;ELSD参数：漂移管温度75℃,喷雾管温度40℃,气体流量1.5L·min-1,输出压力0.5 MPa。结果黄芪甲苷在0.03~1.22mg·mL-1范围内呈良好的线性关系（r=0.999 5）,平均回收率为99.8%,方法精密度的RSD为2.1%（n=6）。结论该方法简单、准确、重复性好,可用于黄芪桂枝五物汤的质量控制。Objective To establish an HPLC-ELSD method for determination of astragalosideⅣin Huangqi Guizhi Wuwu Decoction.Methods The astragalosideⅣin the decoction was determined by HPLC-ELSD.The experiment conditions were as follows：Welchrom-C18（250mm×4.6mm,5μm）column and a mixture of acetonitrile-water（32∶68）as mobile phase.The flow rate was 1.0mL·min-1 and the temperature of drift tube for measurement was 75℃.The pressure of air was 0.5MPa and the flow rate of carrier gas was 1.5L·min-1.Results There was a good linearity within the range of 0.03-1.22mg·mL-1（r= 0.999 5）for astragalosideⅣ,and the average recovery was 99.8% with RSD of 2.1%.Conclusion The method is simple,accurate and reproducible.It can be used as the quality control method of Huangqi Guizhi Wuwu Decoction.
