摘 要:目的建立同时测定金钱草和广金钱草中绿原酸、槲皮素和山柰酚含量的RP-HPLC方法。方法色谱柱为Dikma Diamonsil TMC18柱(250mm×4.6mm,5μm);流动相为甲醇-4mL·L-1甲酸,梯度洗脱;流速为1.0mL·min-1;检测波长360nm。结果绿原酸、槲皮素和山柰酚的线性范围分别为42.08~84.16ng(r=0.999 8),19.13~382.68ng(r=0.999 5)和16.96~339.20ng(r=0.999 5),平均加样回收率分别为99.91%,97.92%和99.31%,方法精密度RSD分别为1.6%,1.4%和1.6%(n=6)。结论该方法快速、简便、准确度高、重复性好、专属性强,可用于金钱草和广金钱草的质量控制。Objective To simultaneously determine chlorogenic acid,quercetin and kaempferol in Herba lysimachiae and desmodium styracifolium,an RP-HPLC method was developed.Methods The separation was performed on a C18(250mm×4.6mm,5μm)column with a gradient elution system of methanol and 4mL·L-1 formic acid at the flow rate of 1.0mL·min-1.The detection wave length was set at 360nm.Results The linear ranges of determination for chlorogenic acid,quercetin and kaempferol were 42.08-84.16ng(r=0.999 8),19.13-382.68ng(r=0.999 5)and 16.96-339.20ng(r=0.999 5),respectively.The average recoveries were 99.91%,97.92% and 99.31%,respectively.The relative standard deviation of precision of the method were 1.6%,1.4%and 1.6%(n=6),respectively.Conclusion The method is rapid,simple,and with high accuracy,good repeatability,strong specificity,and can be used for the Herba lysimachiae and Desmodium styracifolium quality monitoring.
