摘  要:[目的]优化金花茶的离体再生体系。[方法]通过研究不同浓度的ABA、肌醇、琼脂、蔗糖组合及不同光质等方面对金花茶体胚成熟的影响。[结果]在经过ABA的单因素处理后,ABA在浓度为5.0 mg/L时诱导效果最好。在不同浓度的ABA、肌醇、琼脂、蔗糖组合培养基中,诱导体胚成熟的最佳组合为:ABA 10 mg/L+肌醇100 mg/L+蔗糖50 g/L+琼脂6 g/L。在红光、蓝光、绿光、白光培养后,发现只有在白光下诱导的体胚成熟,而在其他单质光中培养的体胚极少成熟。[结论]ABA对金花茶体胚成熟有促进作用,金花茶体胚需要在白光培养下才能成熟。[Objective] The aim was to optimizate the in vitro regeneration system of Camellia nitidissima Chi..[Method]In the experiment,the effects of the different concentrations of ABA,the combinations of different concentrations of ABA,inositol,agar and sucrose,the different colors of light on the mature embryos were compared.[Result] The results showed that after treating with ABA of single factor,the effect was the best when ABA was on the concentration of 5.0 mg/L.In the combination of different concentrations of ABA,inositol,agar and sucrose,the best combination was:ABA 10 mg/L + inositol 100 mg/L + sucrose 50 g/L + agar 6 g/L.Treating the somatic embryos with red light,blue light,green light and white light,then grew on the medium of plantlet formation,the somatic embryos turned red only in the white light,but somatic embryos could not turn red when cultured in other elemental lights.[Conclusion] ABA had acceleration to Camellia nitidissima Chi.on the maturation of somatic embryos.White light was a necessary factor for Camellia nitidissima Chi.on the maturation of somatic embryos.
