摘 要:目的观察吡格列酮(PIO)对高糖(HG)培养的大鼠肾小球系膜细胞(MCs)p22phox和p47phox表达的影响,探讨其肾脏保护机制。方法体外培养MCs,分为正常对照(NG)组、HG组及HG+不同浓度PIO组。RT-PCR测定p22phox、p47phox mRNA表达情况,比色法测定超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性、丙二醛(MDA)含量。结果与NG组比较,HG组p22phox、p47phox mRNA表达明显增加,SOD、CAT活力显著降低,MDA含量明显增高(P<0.01);与HG组比较,各PIO干预组上述变化明显受抑制(P<0.05),且呈剂量依赖性。结论 PIO可抑制MCs还原型烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶表达,减轻氧化应激水平,该作用可能与其肾脏保护部分有关。Objective To observe the effects of pioglitazone (PIO) on the expressions of p22phox and p47phox mRNA in cultured rat glomerular mesangial cells (MCs) and explore its potential reno-protective mechanisms. Methods MCs were cultured in the medium with normal glucose concentration( group NG) , high glucose concentration( group HG) and different concentrations of PIO. The expressions of p22phox and p47phox mRNA were detected by semiquantitative RT-PCR. The levels of superoxide dismutase( SOD), eatalese(CAT) and maleic dialde- hyde(MDA) were determined respectively. Results Compared with group NG, the expressions of p22phox and p47phox mRNA increased significantly, SOD and CAT decreased in group HG, while MDA greatly increased(P 〈 0. 01). When MCs were cultured with PIO, changes mentioned above were improved in a concentration-dependent manner (P 〈 0. 05 ). Conlusion PIO can alleviate high glucose-induced NADPH oxidase expression, lower the oxidative stress in a concentration-dependent manner on MCs, which may partly contribute to its reno-protection.
