摘 要:目的探讨虫草菌粉对兔矽肺纤维化形成过程中的影响。方法选取30只健康雄性大耳白兔随机均分为空白对照组、矽肺对照组、实验干预组。实验干预组予以虫草菌粉喂饮,而空白对照组、矽肺对照组则予以洁净自来水喂饮;2周后矽肺对照组、实验干预组气管内注入二氧化硅粉尘悬液(120 mg/kg),空白对照组气管内注入相同体积的生理盐水。兔染矽尘30 d后处死,计算肺重系数,右肺组织行HE染色,用加碱水解法测定兔肺组织的羟脯氨酸(Hyp)含量,实时荧光定量PCR法测兔肺组织转化生长因子-β1(TGF-β1)及Smad7的mRNA表达水平。结果 3组兔肺组织HE染色显示:空白对照组正常肺组织;矽肺对照组、实验干预组兔肺组织明显纤维化并有矽肺结节形成,显示矽肺兔模型造模成功。在3组兔肺组织中,肺重系数和Hyp的含量比较差异有统计学意义(P<0.05)。与空白对照组比较,实验干预组兔肺组织TGF-β1表达水平升高及Smad7表达水平降低(P<0.05);与矽肺对照组比较,实验干预组兔肺组织TGF-β1表达水平降低及Smad7表达水平升高(P<0.05)。结论虫草菌粉在兔矽肺纤维形成的过程具有一定的影响,可能是通过减少TGF-β1 mRNA表达,依赖于TGF-β-Smads信号通路来调节Smad7 mRNA上调,从而对兔矽肺纤维化的形成有一定抑制作用。Objective To investigate the intervention effects of cordyceps sineses on rabbit silieotic fibrosis model and succeed in creating rabbit silicotic model. Methods Thirty healthy male rabbits were randomly divided into 3 groups : control of blank group, control of silicotic model group, experimental group. The rabbits of control of silicotic model group and experimental group groups were perfused with the mix liquid of silicon dioxide (120 mg/kg) endo- tracheally, and control of blank group with normal saline instead on the same conditions. All the rabbits were sacrificed after 30 days. Calculated Lung/Body coefficient by weighting the lung weight and the body weight of rabbits, the right lung tissue was stained with hematoxylin-eosin (HE). Content of Hydroxyproline was measured by alkaline hydrolysis. The levels of the expression of TGFβ-1 and Smad7 mRNA of rabbits lung sections were measured by Real-time PCR. Results We observed the normal lung tissue in blank group by HE, and the fibrosis and silicotic nodes in the control of silicotic model group and experimental group by HE. The Lung/Body coefficient and the content of Hydroxyproline of rabbits lung tissue were apparently different in the three groups. Compared with the rabbits of control of blank group, TGFβ-1 mRNA of rabbits lung sections were significantly higher and Smad7 mRNA of rabbits lung sections were significantly lower in experimental group; Compared with the rabbits of control of silicotic model group, TGFβ-1 mRNA of rabbits lung sections were significantly lower and Smad7 mRNA of rabbits lung sections were significantly higher in experimental group. Conclusion The cordyceps sinesis may play an important role in the formation of rabbits silicotic fibrosis, and which reduces the expression of TGFβ-1 mRNA and updowns the level of Smad7 by TGFβ-depended Smads signal pathway, which postpones the formation of in rabbit silicotic fibrosis.
