摘 要:目的观察并探讨人滋养细胞表面抗原(Trop-2)在人宫颈病变中的表达及其临床相关性。方法运用RT-PCR法检测6例正常宫颈组织、6例宫颈上皮内瘤变(CIN)Ⅰ或CINⅡ组织、6例CINⅢ组织和6例宫颈癌组织中Trop-2 mRNA的表达情况;免疫组化法检测Trop-2蛋白在20例正常宫颈组织、20例CINⅠ或CINⅡ组织、20例CINⅢ组织及20例宫颈癌组织中的表达情况。结果 Trop-2 mRNA在正常宫颈组织、CINⅠ或CINⅡ组织、CINⅢ组织及宫颈癌组织均有表达,且随着病变程度的加重,表达量呈增加趋势;免疫组化法显示Trop-2蛋白在正常宫颈组织不表达或极低表达,在CIN组织及宫颈癌组织中呈阳性表达,主要定位在细胞膜上,且表达水平随着宫颈病变程度的加重也依次升高。结论 Trop-2在宫颈病变中的表达量和强度与病变的严重程度相关,可以反映宫颈疾病的发生发展过程,有可能成为宫颈疾病筛查及诊断的标记物。Objective To investigate and discuss the expression and the clinical relevance of Trop-2 in human cervical lesions. Methods Reverse transcription-polymerase chain reaction (RT-PCR) and Immunohistochemistry (IHC) were used to detect the expression of Trop-2 in normal cervix, CIN I or CIN Ⅱ , CIN Ⅲ, cervical carcinoma. The expression of Trop-2 protein was detected by IHC in 20 cases respectively and the expresion of Trop-2 mRNA was characterized by RT-PCR in 6 cases respectively. Results RT-PCR showed a expression of Trop-2 in rerespective group and the severer the disease was,the higher mRNA expressed. IHC showed no expression of Trop- 2 protein in normal cervical, whereas, in CIN I or CIN Ⅱ , CINⅢ, cervical carcinoma, the expression of Trop-2 was higher and higher,which was located on the cell membrane. Conclusion The expression of Trop-2 is linked to the occurrence and development of cervix cancer. In addition to a clinical prognostic marker, Trop-2 also may be a potential therapeutic target for cervix cancer.
