摘 要:结合基因功能分类体系(Gene Ontology,GO)对筛选得到的玉米穗粒腐病差异表达基因片段进行全面生物学功能注释。以抗玉米穗粒腐病的自交系Bt-1为材料,接种串珠镰刀菌(Fusarium moniliforme),对接菌后96 h的玉米苞叶样品以玉米全基因组基因芯片进行大规模筛选,利用分子注释系统(Molecular Annotation System,MAS)对筛选得到的基因片段进行GO分子功能注释,分析特征基因的功能。结果表明,利用基因芯片从抗性材料Bt-1中获得的482条差异代表序列,GO注释获得大量重要信息,对应的基因注释包括分子功能、生物过程和细胞组分3个层次。GO能够准确预测与玉米穗粒腐病相关抗病基因的功能,对于了解表达基因之间的相互关系和深入挖掘、理解高通量数据等方面的具有重大帮助,是研究抗病基因不可或缺的生物学信息手段。By Gene ontology(GO),the information of differentially expressed genes after Fusarium moniliforme(FM) infection in maize ear rot was studied.Maize inbred line Bt-1 was use in the experimental material.After inoculating at 96h with FM,treated samples and mock-inoculation controls were investigated by using the genechip maize genome array for hybridization.Each of differentially expressed genes was deeply analyzed by gene ontology annotation.In total,there were 482 unique genes differentially expressed more than 1.5 fold in inbred Bt-1 when compared to mock-inoculated bract tissues from genechip data.Many genes identified by genechip were associated with roles in disease defense for its direct or indirect antifungal activities in response to FM infection.These large responsive genes with significant differentially change were sorted into cellular component,molecular function and biological process with complicated network by molecular annotation system analysis.Altogether,the present Gene ontology analysis provides another view for understanding the molecular mechanism and providing new insights into the deeply complicated network with gene function underlying disease development in the host tissue.
