摘  要:研究高分子顺铂(PGA-Asp-CDDP)腹腔给药昆明鼠,1h后取出脏器检测其铂含量.石墨炉原子吸收光谱测定获得的铂浓度标准曲线,其相关系数为0.997 5,特征浓度为2.4μg·L-1;并考察了消解液用量和消解温度对测定铂浓度的影响;回收率实验表明该消解方法在测定铂浓度上具有可行性和可靠性.在此基础上,昆明鼠的心脏、肝脏、脾脏、肺和肾脏的铂含量检测结果分别为(5.46±0.89)、(28.65±13.45)、(45.30±4.07)、(16.90±4.74)和(76.31±2.59)μg·g-1.The concentrations of Platinum in the heart,liver,spleen,lung,and kidney removed from Kunming mice 1h after they had received intraperitoneal poly(γ-glutamic acid)asp-cisplatin complex(PGA-Asp-CDDP)was determinated.The calibration curve of platinum concentration was obtained by flameless atomic absorption spectrophotometry,the correlation coefficient and the characteristic concentration of the curve were 0.9975and 2.4μg/L.In the study,the effects of experimental conditions,including the amount of acid and digestion temperature on the determination of platinum of PGA-Asp-CDDP were investigated detailedy.Recovery experiment showed that the solubilization method of PGA-Asp-CDDP and tissue was feasible and reliable.On this basis,the Kunming mouse's heart,liver,spleen,lungs and kidneys to be detected,and platinum contents of tissues were(5.46±0.89),(28.65±13.45),(45.30±4.07),(16.90±4.74)and(76.31±2.59)μg/g,respectively.
