摘 要:目的建立反相高效液相色谱法测定人血浆中奥卡西平浓度的方法,为临床合理用药提供依据。方法血浆蛋白经三氯乙酸处理后,采取峰面积外标法,用symmetryC18反相色谱柱,柱温25℃,以乙腈-水(30∶70)为流动相,流速1.0 ml/min,检测波长230 nm。结果血浆中奥卡西平在1.1μg/ml^275μg/ml的浓度范围内线性关系良好,回归方程:Y=56900C+76300.R=0.9998。结论本方法专属性强,灵敏度高,准确度高,线性范围宽,操作简便,分析快速,可用于奥卡西平血药浓度的检测。Objective To establish a RP - HPLC method for the determination of the concentration of Oxcarbazepine in human plasma, in order to provide evidence for the rational drug use in clinic treatment. Methods The plasma samples were simply pretreated by protein precipitation with Trichloroacetic acid. Then the target analyte was separated on symmetry Cls reversed phase column at 25 ℃, with acetonitrile - water( 30: 70) as mobile phase, flow rate of 1.0 ml/min and detection wavelength of 230 nm, and quantified by external standard method. Results During the range from 1.1 μg/ml to 275 μg/ml, it showed upstanding linear relationship between the peak acreage and concentration. The regression equation was Y = 56900C + 76300. Conclusion This method has strong specificity, high sensitivity, high accuracy, wide range of linearity, easy to operate and fast to analyze, it can be used for the determination of Oxcarbazepine in human plasma.
