摘 要:目的以栀子水提取液作为供试样品进行冷冻浓缩和加热蒸发浓缩的对比研究,考察冷冻浓缩技术用于中药水提取液浓缩的可行性。方法取栀子20g,加水10倍、8倍量煮沸2次,每次30min,过滤,合并滤液定容至200mL,测定其糖度。提取液分为2份,每份100mL。第1份样品加热浓缩至50mL;第2份样品预冷至9~10℃,再于-4℃冷冻20min,取出,分离冰晶,另器保存;按上述方法重复至浓缩液体积达50mL时停止冷冻,得冷冻浓缩液;合并冰晶待全部溶解,得冰晶液;分别测定上述两种浓缩液和冰晶液的糖度、栀子苷含量。结果室温浓缩液、冷冻浓缩液、冰晶液糖度分别为4.2°Bx、3.5°Bx、0.2°Bx;栀子苷含量分别为6.530mg/mL、6.461mg/mL、0.52mg/mL;冰晶体积为37.2mL;冷冻浓缩液的可溶性固形物含量高于室温浓缩液,两种浓缩液相对密度和颜色变化不明显,冷冻浓缩液的澄清度好于室温浓缩液。结论采用冷冻浓缩技术对中药有效成分含量和浓缩液性状影响较小,可用于中药水提取液的浓缩。Objective To gardenia extract as samples were frozen concentrated and heated evaporation and concentration of the contrast research, investigation of freeze concentration technology used in water extract of concentrated feasibility. Meth- ods The Gardenia 20g,with 10 times,8 times the amount of boiled two times,each time 30 min,fihering,merging filtrate volume to 200 mL, the sugar content determination. Extract was divided into two parts, each of the 100 mL. The first sample heating concentrated to 50 mL;second samples pre cooled to 9- 10 ℃ DEG C,and then to -4 ~C frozen 20 rain,extrac- tion, separation of ice, the other is saved; according to the above method to stop frozen concentrated liquid product of 50 mL, was frozen concentrate; combined with ice to be completely dissolved, a crystal liquid; the two concentrated liquid and ice crystal liquid were measured in brix, geniposide. Results Room temperature concentrated liquid, frozen concentrated liq- uid, liquid sugar crystals were 4.2° Bx,3.5° Bx,0.2° Bx; geniposide content were 6.530 mg/mL,6.461 mg/mL,0.52 mg/ mL; ice volume 37.2 mL; soluble frozen concentrated liquid solids content is higher than the room temperature liquid con- centrate, two kinds of concentrated liquid relative density and the color change is not obvious, frozen concentrated liquid clar- ification of concentrated liquid at room temperature. Conclusion Use of freeze concentration has little effect on the content of effective components in Chinese medicine and concentrated liquid properties, can be used to concentrate in the water ex- traction liquid.
