左金丸对大鼠应激性胃溃疡保护作用的实验研究

摘　　要：目的:探讨左金丸对因束缚-水浸应激引起的大鼠应激性胃溃疡的影响,并初步探讨其作用机制。方法:将60只健康SD雄性大鼠随机分为6组,即空白对照组,模型对照组,阳性对照组,左金丸低、中、高剂量组,每组10只。各组预防性给药,空白对照组和模型对照大鼠均灌胃生理盐水,其余4组分别灌胃奥美拉唑水悬液,左金丸低、中、高剂量水煎剂。连续灌胃5d。末次给药后禁食不禁水24h,除空白对照组外,其余组均采用"束缚-水浸"应激法制备大鼠应激性胃溃疡模型。造模成功后进行胃体形态学观察,测量胃液PH值,计算胃溃疡指数(UI),并用酶联免疫法检测血清中超氧化物歧化酶(SOD)以及丙二醛(MDA)的含量。结果:左金丸中、高剂量组大鼠胃液PH值明显升高,MDA水平显著下降,左金丸低、中、高剂量组大鼠胃溃疡指数显著降低,血清SOD含量明显上升,与模型对照组比较差异均有统计学意义(P<0.05,P<0.01)。结论:左金丸可以预防应激性胃溃疡的发生并能促进胃溃疡的愈合,其作用机制可能与提高胃液PH值从而降低胃黏膜攻击因素、增强自由基清除能力等因素有关。Objective ：To investigate the effect of Zuojin pills on gastric ulcer caused by water immersion -restraint stress in rats and to preliminarily explore the action mechanism of Zuojin pills. Methods ： Sixty healthy male Sprague - Dawley rats were randomly and equally divided into blank control group ,model control group, positive control group, and low - , middle - , and high -dose Zuojin pill groups. The blank control group and model control group were given normal saline by gavage;the positive control group and low - , mid dle - , and high - dose Zuojin pill groups were intragastrieally administered aqueous suspension of omeprazole and low - , middle - , and high - dose water decoctions of Zuojin pills, respectively. Intragastrie administration was performed for 5 d in each group, followed by 24h of fasting （ without water deprivation）. A rat model of gastric ulcer was established by water immersion - restraint stress in all groups except blank control group. After the model was established, gastric morphology was observed;the pH value of gastric juice was meas ured ; gastric ulcer index （ GUI） was calculated ; serum levels of superoxide dismutase （ SOD ） and malondialdehyde （ MDA ） were deter mined by enzyme -linked immunosorbent assay. Results：The middle - , and high -dose Zuojin pill groups had significantly increased pH value of gastric juice ,and significantly decreased serum MDA level ,the low - ,middle - ,and high -doze Zuojin pill groups had sig nificantly decreased GUI, significantly increased serum SOD level, with significant differences compared with the model control group （P 〈 0. 05 or P 〈 0. 01 ）. Conclusion：Zuojin pills can prevent stress - induced gastric ulcer and promote healing of gastric ulcer, possi bly by increasing the pH value of gastric juice to reduce aggressive factors for the gastric mucosa and improve free radical - scavenging capacity.
