新火针对家兔膝骨关节炎NO、IL-1影响实验研究

摘　　要：目的:观察新火针对家兔膝骨关节炎模型血清一氧化氮(NO)及白细胞介素-1(IL-1)的调节作用。方法:将40只新西兰兔随机分为空白组、模型组、西药组、传统火针组、新火针组,每组8只。采用兔膝关节腔内注入木瓜蛋白酶制造膝骨关节炎模型,造模后第7天开始,分别用玻璃酸钠、传统火针、新火针对各治疗组家兔进行治疗。疗程结束后测定各组血清中NO、IL-1含量。结果:模型组IL-1和NO的含量较空白组升高(P<0.01)。造模后,西药组、传统火针组、新火针组IL-1和NO含量均下降,与模型组比较,差异有显著性或非常显著性意义(P<0.05,P<0.01),提示3种治疗方法均可以改善膝骨关节炎病变时异常升高的IL-1和NO水平。结论:新火针是治疗膝骨关节炎的一种简便有效方法,其作用机制可能是通过降低血清中异常升高的NO、IL-1含量而发挥其抗炎作用,从而延缓软骨的退变。Objective.. To observe the effect of new fire-needling method on nitric oxide（NO） and interleukin-l（IL-1） of the rabbits with experimental knee osteoarthritis（KOA）, and to explore its mechanism. Methods： Forty New Zealand rabbits were equally randomized into blank group, model group, western medicine group, and traditional fire-needling group and new fire-needling group. KOA model was established with reference to KITOH method. Seven days after the modeling, the rabbits in the treatment groups were given sodium hyaluronate（SH）, traditional fire-needling method and new fire-needling method. The serum concentrations of NO and IL-1 were detected by enzyme linked immunosorbent assay（ELISAI method at the end of the course of treatment. Results： In comparison with the blank group, the serum concentrations of NO and IL-1 were in- creased significantly in the model group（P 〈 0.01）. Compared with the model group, the concentrations of NO and IL-1 were lowered significantly in western medicine group, traditional fire-needling group and new fire-needling group after treatment（P 〈 0.05, P 〈 0.01）, indicating that all of the three therapies could improve the NO and IL-1 concentrations. Conclusion. New fire-needling method is a simple and effective method for treatment of knee osteoarthritis, and its anti-inflammatory mecha-nism may be related with the regulation of the abnormal serum NO and IL-1 levels.
