赤芍801诱导类风湿关节炎成纤维样滑膜细胞凋亡研究

摘　　要：目的探讨赤芍801对类风湿关节炎成纤维样滑膜细胞凋亡的影响。方法 RA患者关节滑液经原代培养分离出FLS,MTT检测细胞增殖抑制。根据MTT结果,将细胞分为高剂量赤芍801组(64μg/ml)、低剂量赤芍801组(4μg/ml)和溶剂对照组,倒置显微镜观察其细胞形态;Annexin V-FITC/PI双标记染色检测细胞凋亡率。结果 MTT结果显示,赤芍801具有浓度依赖性的抑制RA FLS增殖功能。流式结果表明,赤芍801具有诱导RA FLS凋亡功能,虽然低剂量组的凋亡率与对照组比较增加不明显(P>0.05),但高剂量组的凋亡率较对照组显著增加(P<0.05)。结论赤芍801具有抑制RA FLS增殖及诱导其凋亡功能。Objective To explore the effect of radix paeoniae rubra 801 （P801） on the apoptosis of fibroblast like syno viocytes （FL,Ss） from patients with rheumatoid arthritis （RA）. Methods RA FLSs were primarily cultured from articular genu synovia of RA patients. Cell proliferation was determined by MTT assay. According to the results of MTT, the cells were divided into high dose group of P801 （64μg/ml）, low- dose group of P801 （4μg/ml） and solvent control group. The cell mor- phology was observed by the optical inverted microscope. And the apoptotie rate was detected by Annexin V- FITC/PI double la- beling staining. Results MTT results showed that P801 had the ability to inhibit RA FLS proliferation with a concentration dependence manner. Flow cytometry results showed that P801 could induce the apoptosis of RA FLS. The apoptotic rate in low dose group of PS01 did not significantly increase as compared with the control （P 〉 0.05）, but it was significantly increased in high dose group of P801 （P 〈0.05）. Conclusions P801 has the function to inhibit proliferation of RA FLS and induce its apoptosis.
