摘 要:为探讨芦笋(Asparagus officinalis L.)玻璃化试管苗的逆转恢复途径,对培养温度、光照强度、蔗糖浓度、琼脂浓度等因素对玻璃化苗逆转效果的影响进行研究。结果显示:玻璃化苗的恢复率随着培养温度的降低而有所升高,培养温度为15℃时,恢复率为33.01%;玻璃化苗的恢复率随光照强度的增强而明显升高,当光照强度为5200 lx时,逆转率可达70.55%;玻璃化苗的恢复率随着蔗糖浓度的升高而逐渐升高,当蔗糖浓度为50 g/L时,恢复率可达48.20%;玻璃化苗的恢复率随琼脂浓度的增加而升高,琼脂浓度为9 g/L时,恢复率达38.18%。说明在4种影响因素中,光照强度对玻璃化苗逆转的影响最大,而培养温度、蔗糖浓度、琼脂浓度对玻璃化苗的逆转也有一定的影响。The reversion of vitrification of Asparagus of6cinalis L. during tissue culture was discussed. The influence factors on the reversion of vitrification were studied,, including culture temperatures, culture illumination, concentration of sucrose and agar. The results showed that the percentage of the reversion of vitrification would be increased with decreasing temperature, the rate of reversion was 33.01%, when the culture temperature at 15℃. The rate of reversion would be increased obviously along with the increase of culture illumination, the rate would amount to 70.55%, under the., intensity of 5200 lx culture illumination. With the increase of sucrose concentration, the recovery rate increased gradually, when the sucrose concentration was 50 g/L, the recovery rate increased to 48.20%. The rate of reversion increased with the agar concentration increasing, when the agar concentration was 9 g/L, the recovery rate was 38.18%. The culture illumination showed the best effect on the reversion of vitrication in the 4 factors. Moreover, the culture temperatures, the concentration of sucrose and agar showed some efficacious effects on the reversion of vitrication.
