摘 要:目的本研究旨在探索儿童再生障碍性贫血(AA)骨髓间充质干细胞(MSC)GATA-2基因的表达及意义。方法采集38例AA患儿骨髓标本及20例正常儿童骨髓标本,分离骨髓单个核细胞后体外扩增培养骨髓基质细胞,实时定量PCR(QRT-PCR)方法检测AA儿童免疫抑制治疗前后GATA-2基因表达的变化。结果 AA儿童免疫抑制治疗前GATA-2基因表达水平明显低于正常对照组(P<0.05),免疫抑制治疗2年后对治疗有反应的AA儿童GATA-2表达水平高于发病时,且与正常对照组表达水平无统计学差异(P>0.05)。结论 GATA-2基因的低表达可能影响AA患儿骨髓微环境的造血调控作用。Objective To investigate the expression of transcription factor GATA-2 genes in the bone marrow MSCs from children with aplastic anemia (AA) and normal controls. Methods We collected bone marrow samples from thir ty-eight aplastic anemia patients and normal persons,The expression of GATA-2 genes were analyzed by using QRT- PCR in the MSCs from 20 normal controls and 38 cases with AA. The expression levels of GATA-2 genes in the MSCs from children with AA were compared before and after therapy. Results GATA-2 expression by MSCs from AA pa tients at diagnosis was significantly lower than in normal controls (p〈0.05). After two years of immunosuppressive therapy fIST), expression of GATA-2 from responders was significantly higher than those at diagnosis. It was no difference in the expression level of GATA-2 gene within the MSCs from normal controls and responders. Conclusion The lower expression of GATA-2 in the MSCs from children with AA may influence the regulation of hematopoiesis and play the role in pathogenesis of AA.
