摘  要:目的研究促红细胞生成素(EPO)对顺铂(CDDP)诱导的人胚肾细胞(HEK-293)毒性的影响及其可能机理。方法体外培养HEK-293细胞,采用MTT法检测不同浓度CDDP、EPO对HEK-293细胞的作用,倒置显微镜观察细胞形态变化,硫代巴比妥酸法检测细胞丙二醛(MDA)含量,比色法测定超氧化物歧化酶(SOD)活性变化。结果 CDDP能明显抑制HEK-293细胞存活率,诱导细胞贴壁能力下降,增加MDA含量,降低SOD活性;而加入EPO后能明显改善这些状况。结论 EPO对CDDP诱导的HEK-293细胞损伤具有保护作用。其保护机制可能与其抗氧化活性、猝灭自由基的功能有关。Objective To investigate the protective effect and underlying mechanism of erythropoietin(EPO) on cisplatin(CDDP)-induced cytotoxicity in human embryo kidney-293 cells (HEK-293 cells). Methods HEK-293 cells were cultured in vitro, and the effects of different concentrations of EPO and CDDP on HEK-293 cells were evaluated by MTT assay. The changes of cell morphology were observed under inverted microscope. The content of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) were detected by thiobarbituricacid and colorimetry methods, respectively. Results CDDP could significantly inhibit survival rate of HEK-293 cells, decrease anchorage-dependent cells, upregulate MDA content and downregulate SOD activity, which were all improved by EPO treatment. Conclusion EPO has a protective effect on CDDP-induced cytotoxicity in HEK-293 cells, which may be related with antioxidation and scavenging free radicals.
