摘 要:为分析广西猪圆环病毒2型(PCV2)流行毒株的类型和特点,为今后预测PCV2进化、探讨合适的防治对策提供理论指导,对广西不同市(县)送检的猪脾、肺、肾、淋巴结及流产胎儿样本进行了PCV2检测及基因克隆和序列分析,并应用重组软件RDP3.0、SimPlot3.5.1软件和BOOTSCAN分别对所得基因序列进行了基因重组情况、相似性和遗传进化分析。结果显示,成功克隆了22株PCV2全基因序列,其中有4株属于PCV2d型,1株属于PCV2e型,其余的为PCV2b型。检测到2个重组序列(GXNN-5,GXNN-7),都归属于PCV2b型,重组都发生在ORF1基因区,重组片段NJ进化树的拓扑结构也发生了变化。证实,广西目前流行的PCV2以PCV2b和PCV2d为多,其中以PCV2b型为主。存在两毒株重组现象,重组亲本序列很可能来源于PCV2b和PCV2d型。This research intended to analyze the strains type and characteristics of porcine circovirus type 2(PCV2) ,and to provide a theoretical guidance for the future evolution of forecast PCV2 and explore suitable control measures. The spleen,lung,kidney, lymphonedes and aborted fetus samples from different counties of Guangxi were tested for PCV2. Their genome sequences were cloned, sequenced and then ana- lyzed. RDP3.0, SimPlot3.5.1 and BOOTSCAN softwares were used to analyze the similarity of the recom- binants and their genetic evolution. Twenty-two complete genome sequences were cloned,in which 4 of the strains obtained in this study belonged to PCV2d,1 belonged to PCV2e and others belonged to PCV2b. The 2 putative recombinants obtained named GXNN-5 and GXNN-7 that belonged to PCV2b were detected with RDP3.0,and SimPlot3.5.1 softwares. The recombination occurred in ORF1 region,and changes in topolo- gy of phylogenetic tree were detected with Neighbor-Joining(NJ) method. This study proved that recently PCV2(PCV2b and PCV2d sub-genotypes) was the most popular in Guangxi with a high prevalence of PCV2b. The existing recombination phenomenon and their parental relationship may have originated from PCV2b and PCV2d sub-genotypes.
