摘  要:目的:研究白细胞介素8(IL-8)是否能够诱导心脏成纤维细胞(CF)的迁移运动,并验证该过程是否涉及丝裂原活化蛋白激酶(MAPK)亚家族ERK1/2、P38、c-JNK信号转导通路。方法:应用Transwell技术研究IL-8对CF的趋化诱导作用,以及MAPK信号转导通路与这种迁移的关系。结果:①IL-8在10-8~10-10 g/ml时均能诱导CF迁移运动,并在10-9 g/ml时迁移效应最明显;②IL-8诱导CF的迁移作用开始于4~8h,并呈现时间依赖性;③MAPK亚家族ERK1/2、P38、c-JNK信号转导通路在IL-8诱导的CF迁移过程中均发挥作用。结论:IL-8能够诱导CF迁移运动,且这种迁移过程可能有MAPK亚家族ERK1/2、P38、c-JNK信号转导通路参与。Objective:To test weather interleukin-8 (IL-8) could induce the cardiac fibroblasts (CF) migration and mitogen-activated protein kinase (MAPK) signaling pathways including ERK1/2, P38, c-JNK were involved in the migration process. Method= The migration and effect of the MAPK signaling pathways involved were esti- mated by transwell chamber in vitro. Result= (I)IL-8 stimulated the migration function of CF at the concentration range of 10 8 g/ml to 10 10 g/ml, especially at 10 ^-9g/ml. (2)The migration began at 4--8 h with a dose- dependent manner. (3)All three MAPK pathways including ERK1/2, P38, c-JNK took effect on the migration process. Conclusion:IL-8 could induce CF migration via all three MAPK signaling pathways including ERK1/2, P38, and c-JNK.
