摘  要:目的:探讨吸烟对大鼠心肌组织自噬相关基因及蛋白表达的影响。方法:雄性SD大鼠20只,随机分为吸烟组和对照组,建立吸烟模型,并用RT-PCR方法检测两组心肌组织中Beclin-1和LC3ⅡmRNA的表达,用Western Blot方法检测相应蛋白的表达水平。结果:吸烟组自噬基因Beclin-1(0.794±0.169)较对照组Beclin-1(0.402±0.087)表达增加(P<0.01),吸烟组自噬基因LC3Ⅱ(0.684±0.107)较对照组(0.290±0.084)表达增加(P<0.01),相应蛋白吸烟组Beclin-1(1.304±0.587)、LC3Ⅱ(10.924±2.622)表达较对照组Beclin-1(0.143±0.051)、LC3Ⅱ(4.361±1.164)表达增加(P<0.01)。结论:吸烟可引起大鼠心肌组织自噬相关基因Beclin-1和LC3Ⅱ及相应蛋白表达增加。Objective: To investigate the effects of cigarette smoking on the expression of autophagy related genes in rat myoeardium. Method:Twenty male SD rats were randomly divided into control group (n= 10) and cig- arette smoking (CS) group (n: 10). The mRNA and protein expression of autophagy related genes were deter- mined by RT-PCR and Western Blot, respectively. Result: The Beclin-1 mRNA expression was significantly increased in CS group (0.794_±0.169) compared to the control group (0.402±0.087) (P〈0.01), and the pro- tein expression of Beclin 1 was also higher in the CS group than that in control (1. 304±0. 587 vs 0. 143±0. 051, P〈0.01). Accordingly, the CS group showed significantly higher level of LC3 II mRNA (0. 684±0. 107) com- pared to the control group (0.290±0.084, P〈0.01), as well as the LC3II protein expression (10.924±2.622 vs 4. 361±1.164, P〈0.01). Conclusion:Cigarette smoking up-regulates the mRNA and protein expression of autophagy related genes in rat myocardium.
