摘  要:目的 探讨利鲁唑(riluzole)对慢性背根节压迫(chronic compression of dorsal root ganglion,CCD)模型大鼠背根节(dorsal root ganglion,DRG)神经元诱发簇放电(evoked-bursting,EB)的抑制作用及电流机制.方法 33只健康SD大鼠,随机分为正常对照组(n=13)及CCD模型组(n=20),进行离体膜片钳全细胞记录.结果 CCD术后DRG神经元EB发生率增高,达53%(44/83),与正常对照组比较差异有统计学意义(P〈0.05);DRG局部给予利鲁唑(1、10、100 μmol/L)浓度依赖性抑制持续性钠电流(persistent sodium current,INaP),100 μmol/L利鲁唑可抑制阈下膜电位振荡幅度及EB放电.结论 利鲁唑通过阻断INaP抑制EB放电而发挥外周镇痛作用.Objective To investigate the inhibitory effect and current mechanism of riluzole on evoked - bursting (EB) firing in dorsal root ganglion (DRG) neurons in chronic compression of the DRG (CCD) model rats. Methods 33 SD rats were randomly divided into normal (n = 13) and CCD model (n =20) groups. In vitro patch clamp recording was done on DRG neurons. Results In CCD group the incidence of EB firing increased significantly compared to normal group (P 〈0.05),EB was inhibited in a reversible way by riluzole concentration dependently (l, 10, 100 ptmol/L) through persistent sodium current (INaP) , which participated in subthreshold membrane potential oscillation (SMPO) for- mation. Conclusion Riluzole might play analgesic effect in peripheral nervous system by selectively inhibiting INaP.
