摘  要:目的探讨酸性成纤维细胞因子(aFGF)对新生大鼠海马星形胶质细胞生长的影响及其对庆大霉素(GM)损伤海马星形胶质细胞的保护作用。方法取新生乳鼠,分离海马、剪切、胰蛋白酶消化,制成细胞悬液后接种。采用GTAP免疫组化染色鉴定。传3代细胞接种于96孔培养板或培养瓶:(1)培养24 h后加入一系列浓度的aFGF,再培养24 h后用MTT法检测细胞存活率;(2)GM组:2 g/L GM作用24 h;aFGF+GM组:加入一系列浓度aFGF 24h后再加2 g/L GM培养24 h,MTT法检测细胞存活率;观察细胞超微结构变化。结果星形胶质细胞在aFGF作用下有不同程度的增殖,并在一定范围内呈浓度依赖性(P﹤0.01);不同浓度的aFGF预作用24 h后,使GM致伤的星形胶质细胞存活率升高,并能减轻星形胶质细胞超微结构的破坏。结论 aFGF能促进大鼠海马星形胶质细胞的增殖及拮抗GM对星形胶质细胞的损伤作用。OBJECTIVE The role of acidic fibroblast growth factor (aFGF) on the growth of hippoeampal astrocytes in neonatal rats and its protective effect against hippocampal astroeyte damage caused by gentamicin (GM) were investigated. METHODS Hippoeampus of lactating rats was removed, sheared, and trypsinized to a cell suspension for inoculation. GTAP immunohistochemical analysis was used in this study. Cells at 3rd passage were cultured in a 96-well plate or flask : ( 1 ) After 24 h of incubation, aFGF of different concentrations was added to the culture, followed by an additional 24 h of incubation. MTT assay was then performed to determine cell viability of the samples. (2) GM group: 2g/L of GM was added to the cultures and incubated for 24 h ; aFGF+GM group: aFGF of different concentrations was added to the culture and incubated for 24 h. 2g/L of GM was then added to the culture and incubated for another 24 h. MTT assay was used to determine cell viability of the samples. Changes in ultrastructure of the cell samples were observed. RESULTS Astrocytes exhibited a dosedependent proliferation in the presence of aFGF (P 〈 0.01) ; pre-incubation with different concentrations of aFGF for 24 h improved the survival rate of GM-damaged astrocytes and reduced the level of damage in astrocyte uhrastructures. CONCLU-SION aFGF has proliferative effects on rat hippocampal astrocytes and may inhibit the damaging effect of GM on astrocytes.
