摘  要:目的:制定神安颗粒中水溶性成分的指纹图谱,为神安颗粒"全指纹图谱系"的建立提供依据。方法:采用高效液相色谱法,以斯皮诺素为参照物,色谱柱为C18(250 mm×4.6 mm,5μm),流动相为乙腈-水(梯度洗脱),流速为1.0 ml/min,柱温为室温,检测波长为335 nm,进样量为10μl,记录时间为55 min。以斯皮诺素峰(S峰)的保留时间和峰面积为1,计算各色谱峰的相对保留时间和相对峰面积,并进行方法学考察;采用国家药典委员会推荐的"中药色谱指纹图谱相似度评价系统(2004A版)",对10批样品进行相似度评价。结果:制定了神安颗粒中水溶性成分的区间指纹图谱,标注了6个共有指纹峰。各共有指纹峰的相对保留时间和相对峰面积基本一致,RSD<2%;10批样品的相似度均在0.80以上。结论:通过对神安颗粒中水溶性成分指纹图谱的制定,可为其"全指纹图谱系"的建立提供试验依据,为提高天然产物质量控制技术进行有意义的探索。OBJECTIVE:To establish fingerprint of water-soluble components of Shen'an granules,and to provide reference for the establishment of"complete fingerprint system"of Shen'an granules.METHODS:HPLC method was adopted using spinosin as reference.C18(250 mm×4.6 mm,5 μm)column was used with acetonitrile-water as mobile phase(gradient elution)at the flow rate of 1.0 ml/min.The column temperature was room temperature,and detection wavelength was set at 335 nm.The sample size was 10μl,and record time was 55 min.The retention time and peak area of spinosin chromatographic peak(S peak)regarded as 1,relative retention time and relative peak area ratios of other peak were calculated and reviewed by methodology;the similarity of 10 batches of samples was evaluated by using"TCM chromatograph fingerprint similarity appreciation system"recommended by Chinese Pharmacopoeia Committee.RESULTS:The interval fingerprint of water-soluble constituent of Shen'an granules had been formulated,and 6 common peaks were marked.The relative retention time and relative peak area of each common peak were in substantial agreement(RSD2%);10 batches of samples showed the similarity was all more than 0.80.CONCLUSIONS:The formulation of the fingerprint of water-soluble constituent of Shen'an granules provide experiment basis for the construction of"complete fingerprint system",and carry out significant exploration to improve quality controlling technology of natural products.
