氟诱发Runx2高表达对成纤维细胞成骨作用的影响

摘  要:目的通过观察氟致大鼠皮肤成纤维细胞中核心结合因子(Runx2)浓度及碱性磷酸酶(ALP)活力的变化,探究氟对成纤维细胞成骨作用的影响。方法选取和培养大鼠皮肤成纤维细胞,分7个染氟组(染氟浓度分别为0.0001、0.001、0.01、0.1、1.0、10、20mg/L)和对照组,检测和分析染氟后不同时间(24、48、72、96h)各组培养液中Runx2浓度和ALP活力。结果 72h时0.001、0.1、1、10、20mg/L染氟浓度组及96h时所有染氟组Runx2的浓度较对照组均明显升高,差异有统计学意义(F=17.59,P<0.05;F=13.31,P<0.05)。72h时0.1、20mg/L浓度组及96h时0.001、0.1、10、20mg/L浓度组ALP活力较对照组明显升高,差异有统计学意义(F=30.05,P<0.05;F=27.59,P<0.05)。结论氟暴露致成纤维细胞中Runx2浓度升高,并且ALP活力增强。提示,氟可诱发成纤维细胞Runx2的高表达,以此促进成纤维细胞的成骨作用。Objective To explore the influence of fluoride on ossification of fibroblast through observing the expression changes of Runx2 and ALP on fibroblast under fluoride exposure. Methods The skin fibroblasts of adult rat were divided into 7 fluoride exposed groups(0. 0001,0. 001,0. 01,0. 1,1.0, 10 and 20 mg/L)and a control group. The levers of Runx2 and ALP in every group were analyzed at different time periods(24, 48, 72, and 96 h). Results The concentrations of Runx2 in different doses and time periods showed that the concentrations of Runx2 in groups of 0. 001, 0.1, 1, 10 and 20 mg/L at 72 h and all the groups at 96h were higher than that in control group, the differences were significant(F= 17.59, P〈0. 05;F= 13.31 ,P〈0. 05). The activity levels of ALP in different doses and time periods showed that the activities of ALP in groups of 0. 1 and 20 mg/L at 72 h and 0. 001,0. 1,10 and 20 mg/L at 96 h were higher than that in control group, the differences were significant (F = 30.05, P 〈 0.05; F = 27.59, P 〈 0.05). Conclusions Fluoride exposure may cause the overexpression of Runx2 accompanied with the increase of ALP activity. It suggests that fluoride could induce the overexpression of Runx2 so as to promote the ossification of fibroblast.
