摘 要:目的探讨补肾生血解毒方对理化复合因素致骨髓抑制小鼠造血微环境中造血干细胞增殖的影响。方法 BALB/C小鼠随机分为空白组,模型组,中药治疗小、中、大剂量组,以^(60)Co-γ射线复合环磷酰胺致小鼠骨髓造血抑制为动物模型,补肾生血解毒方(龟板胶、鹿角胶、人参、枸杞子、黄芪、当归、黄连)干预,中药小、中、大剂量组每天分别给予相当于生药量6.14、12.29、24.57 g/kg。流式细胞仪检测小鼠骨髓细胞中CD45^+CD117^+细胞的数量,酶联免疫吸附测定(ELISA)检测外周血血清中干细胞因子(SCF)含量的差异,RT-PCR检测骨髓细胞中干细胞因子受体(c-kit)基因表达的改变。结果该方可以显著增加小鼠骨髓细胞中CD45^+CD117^+造血干细胞数量,与模型组相比差异有显著性(P<0.01);外周血血清中SCF含量显著高于空白组和模型组(P<0.01),空白组和模型组之间无明显差异(P>0.05);各组小鼠骨髓细胞中c-kit基因表达无显著性差异(P>0.05),中药治疗组与模型组相比有升高趋势。结论补肾生血解毒方治疗再障可能与提高SCF活性和诱导造血干细胞增殖、分化有关,这也是该方疗效的主要作用机制之一。Objective To investigate the influence of Bushen Shengxue Jiedu Fang on proliferation of hemopoietic stem cells (HSCs) in mice with myelosuppression induced by physiochemical compound factors.Methods BALB/C mice were randomly divided into blank group,model group,high-dose,mid-dose and low-dose Bushen Shengxue Jiedu Fang groups (high-dose group,mid-dose group and low-dose group).The mouse model of myelosuppression was established with 60Co-γay and cyclophosphamide.The high-dose group,mid-dose group and low-dose group were given,respectively,Bushen Shengxue Jiedu Fang in the dose of 6.14 g/kg,12.29 g/kg and 24.57 g/kg (equal to crude medicinal quantity) every day.The quantity of CD45 + CD117+ in HSCs was detected by using flow cytometer,difference in content of serum stem cell factor (SCF) was detected by using enzyme-linked immunosorbent assay (ELISA) and the expression of c-kit gene was detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR).Results The quantity of CD45 + CD117 + increased significantly in Bushen Shengxue Jiedu Fang groups compared with model group (P <0.01).The content of serum SCF was significantly higher in Bushen Shengxue Jiedu Fang groups than that in blank group and model group,but there was no significant difference between blank group and model group (P <0.01).The expression of c-kit gene had no significant difference among all groups,while had increasing tendency in Bushen Shengxue Jiedu Fang groups compared with model group.Conclusion The curative effect of Bushen Shengxue Jiedu Fang on aplastic anemia may be related to that it can promote SCF activity and induce the proliferation and differentiation of HSCs,which is one of key mechanisms of its curative effect.
