摘 要:目的 建立参七消痞颗粒中野黄芩苷和丹酚酸B的含量测定方法.方法 采用HPLC法,色谱柱为Waters SunFireTM C18(4.6×150 mm,5μm),流动相为甲醇-0.1%磷酸水溶液梯度洗脱,流速为1.0 mL/min,柱温为30℃,野黄芩苷和丹酚酸B检测波长分别为335 nm、280 nm.结果 野黄芩苷和丹酚酸B分别在0.064 16 ~0.385 0μg和0.743 ~4.455 μg质量范围内与色谱峰面积呈良好线性关系,相关系数分别为0.999 5和0.999 4.野黄芩苷和丹酚酸B的平均加样回收率分别为101.9%和96.3%,RSD分别为1.17%和0.33%.结论 本方法简单可靠,结果准确,可用于参七消痞颗粒的质量控制.Objective To establish a method for quantitative determination of scutellarin and salvianolic acid B in Shenqi Xiaopi Granules.Methods The HPLC procedure was performed on the chromatographic column of Waters SunFireTM C18 (4.6 x 150 mm,5 μm) at 30℃,and the mobile phase was methanol-0.1% phosphoric acid solution in gradient elution.The flow velocity was 1.0 mL/min.The detection wavelength of scutellarin was 335 nm,and that of salvianolic acid B was 280 nm.Results Scutellarin and salvianolic acid B respectively showed a good linear relationship with peak area at a range from 0.064 16 μg to 0.385 0 μg and from 0.743 μg to 4.455 μg,and the correlation coefficients were 0.999 5 and 0.999 4.The average recovery of scutellarin was 101.9 % (RSD =1.17%),and that of salvianolic acid B was 96.3 % (RSD =0.33%).Conclusion The method is simple,reliable and accurate,which can be used for the quality control of Shenqi Xiaopi Granules.
