摘 要:利用基于16SrDNA的PCR-DGGE技术结合特异性和共性条带割胶回收DNA进行克隆和测序,监测了饲喂不同剂量重组人溶菌酶(rhLYZ)后小鼠在不同时间内粪样细菌区系以及菌群结构多样性的变化,并鉴定了部分特异性和共性菌群成员,分析rhLYZ对小鼠肠道微生物群落结构和多样性的影响。结果显示,随着小鼠日龄的增加,其粪样微生物种类趋于多样性。在口服接种rhLYZ后,与对照组相比,除高剂量组(1g/L)小鼠粪样微生物多样性较对照组明显减少外,中剂量(0.5g/L)、低剂量(0.25g/L)及极低剂量组(0.10g/L)组差异不显著。结果表明,rhLYZ对小鼠肠道菌群结构影响作用呈现剂量依赖性。与对照组相比,各接种剂量组小鼠肠道中优势菌种保持不变,主要有拟杆菌、马胃葡萄球菌、缓慢葡萄球菌等葡萄球菌属,约乳杆菌、罗伊乳杆菌等乳酸菌属。The bacterial community and diversity in the mice intestinal tract were studied by using 16S rDNA based denaturing gradient gel electrophoresis(DGGE)and some common and special bands were identified,in order to investigate the effect of different concentration dose of rhLYZ on the mice intestinal flora structure by feeding.The results showed that the microbes of all the mice feces had an increasing trend as the mice growed.After feeding the mice with rhLYZ,the microbial diversity of the high-dose group(1g/L)significantly reduced(P0.05)while the medium-dose group(0.5g/L),the low-dose group(0.25g/L)and the lowest-dose group had no significant difference(P0.01)compared with the control group,which indicated that the effect of feeding rhLYZ on animal intestinal bacterial community and diversity was dose dependent.In addition,the predominant bacterial of mice intestinal tract in each dose group remain unchanged,which mainly contained Bacteroides sp.,Staphylococcus equorumand Staphylococcus lentus,Lactobacillus johnsonii strain and Lactobacillus reuteri strain.
