猪IL-18在猪繁殖与呼吸综合征灭活疫苗中的免疫佐剂作用

摘　　要：为了探讨猪IL-18对猪繁殖与呼吸综合征灭活苗体液免疫和细胞免疫的影响,75头21日龄断奶仔猪随机分成5组,将重组猪IL-18蛋白、pcDNA3/pmIL-18质粒均分别以200μg/次和400μg/次剂量与猪繁殖与呼吸综合征灭活苗同时免疫,于免疫后7、15、30、45、60d采集血液,以MTT法检测猪外周血淋巴细胞增殖情况,以流式细胞术检测猪外周血中CD4+和CD8+的变化,以ELISA法检测免疫猪猪繁殖与呼吸综合征抗体变化。结果显示,猪IL-18能够显著促进猪外周淋巴细胞增殖,可明显提高猪繁殖与呼吸综合征灭活苗免疫猪外周血中CD4+和CD8+的数量,无论是重组猪IL-18蛋白还是pcDNA3/pmIL-18质粒均可提高PRRSV灭活苗抗体水平,且重组猪IL-18高、低剂量组在免疫30d猪体中猪繁殖与呼吸综合征灭活苗的抗体水平达到高峰,随后下降,剂量越高提高作用越明显;而pcDNA3/pmIL-18高、低剂量组在免疫45d猪体中猪繁殖与呼吸综合征灭活苗的抗体水平达到高峰后下降,剂量越高促进作用越好。猪IL-18可作为猪繁殖与呼吸综合征灭活苗的免疫佐剂。To study the effects of porcine interleukin 18 on humoral and cellular immunity of pig to porcine reproductive and respiratory syndrome（PRRS）inactivated vaccine. 75 heads of 21-days-old weanling piglets divided randomly into five groups. Two groups were vaccinated respectively with recombinant in- terleukin 18 （IL-18） and PRRS inactivated vaccine together at 200 μg or 400 μg once. Another two groups also were vaccinated respectively with pcDNA3/pmIL-18 plasmid and PRRS inactivated vaccine together at 200μg or 400 μg once. The control group was vaccinated with physiological saline and PRRS inactivated vaccine at the same dose. 7,15,30,45,60 days after immunization, peripheral blood was collected for de- tection of peripheral lymphocyte by MTT,CD4＋ and CD8＋ T lymphocytes were examined by flow cytome- try and the serum were collected to examine antibodies to PRRS inactivated vaccine by ELISA. The results showed that porcine IL-18 has obvious lymphocyte proliferation, and also it can increase amount of CD4＋ and CD8＋ T lymphocytes in the peripheral blood of piglets after immunization. Both recombinant IL-18 or pcDNA3/pmIL-18 plasmid can enhanced antibody titer to PRRS inactivated vaccine. The antibody level reached the peak after vaccination 30 days in two recombinant IL-18 groups and 45 days in two pcDNA3/ pmIL-18 plasmid groups. So porcine IL-18 is a potential immunoadjuvant for PRRS inactivated vaccine
