IL4对肝片吸虫感染诱导巨噬细胞中脂肪滴分布的影响

摘  要:试验以肝片吸虫囊蚴经口分别感染雌性BALB/c野生型及其IL4单抗处置小鼠,运用特异性PCR鉴定成功感染小鼠后获取巨噬细胞,设立人工巯基醋酸盐诱导巨噬细胞对照组进行体外培养。用荧光定量PCR检测巨噬细胞的标记蛋白Relmα和Ym1以确定其激活状态。将所获取的巨噬细胞的细胞质用特异性抗体染色,并用共聚焦显微镜拍摄不同时间点从不同小鼠体内获取的巨噬细胞染色后的图片。结果发现,Relmα和Ym1基因在IL4单抗处置小鼠和巯基醋酸盐诱导小鼠的巨噬细胞中均表达,但在IL4单抗处置小鼠的表达量显著下降。脂肪滴分布在肝片吸虫诱导的选择性激活巨噬细胞的细胞质中,被染色成绿色且呈圆形。同时发现脂肪滴的分布形式多样,大小不一,既有单个分布也能成蔟分布。Macrophages were acquired from the Fasciola hepatica metacercariae infected-, IL4 mAb treated- and wild type female BALB/c mice, respectively. The species-specific PCR was applied to identify the successful infection of FF. hepatica, mRNAs from macrophages were extracted for the detection of Relma and Yml genes using real-time PCR. Pictures at different time points were acquired with a confocal microscopy. Results demonstrated that levels of Relma and Yml in F. hepatica elicited macrophages (F- M2)and thio-elicited macrophages(T-M2) from IL4 mAb treated mice were significantly decreased with comparison to wild type mice. The fat granules were distributed diversely in the cytoplasm of F-M2 cells with a green and round appearance.
