寻常性银屑病患者外周血和皮损中Thl7细胞表达及相关性研究

摘　　要：目的探讨寻常性银屑病患者外周血和皮损中Th17细胞及相关因子IL-17、IL-22表达水平与疾病严重程度的相关性。方法抽取寻常性银屑病患者和正常人外周血,用流式细胞仪检测外周血Thl7细胞,ELISA试剂盒检测血清中IL-17、IL-22表达。取寻常性银屑病患者皮损和正常人对照者皮肤,用量子点免疫荧光双标法检测Thl7细胞表达。结果两组患者外周血中CD4及IL-17抗体阳性表达率相比差异有统计学意义(P<0.05);实验组患者外周血IL-17和IL-23表达水平明显高于对照组(P<0.05),并与PASI呈正相关(r=0.501,P<0.05);寻常性银屑病患者皮损和正常人对照者皮肤组织中Th17细胞表达比例差异有统计学意义(P<0.05)。结论 Th细胞比例在寻常型银屑病患者外周血和皮损中明显增加,IL-17和IL-22的水平均升高。Objective To investigate the correlation between the severity of psoriasis vulgaris and the expression levels of Thl7 cells and related factors,IL-17,IL-22 in peripheral blood and lesions of patients with psoriasis vulgaris.Methods 78 cases of patients with psoriasis vulgaris were selected as the observation group and 48 cases of outpatients and general surgical patients with no psoriasis vulgaris and other immune related diseases were selected as the control group.All the patients＇ Thl7 cells in peripheral blood were tested by flow cytometry and the expressions of IL-17,IL-22 in serum were detected by ELISA kit.And the expressions of Thl7 cells in lesions of the observation group and the skin of the control group were detected by quantum dots immunofluorescence double-labeling method.Results There was significant difference between the positive expression of CD4 and IL-17 antibody in peripheral blood of the two groups（P0.05）;the expression levels of IL-17 and IL-23 in peripheral blood of the observation group were obviously higher than those of the control group（P0.05）,and which were positively correlated with PASI（r=0.501,P0.05）;Th17 cells expression ratio in the observation group＇s lesions was significantly different from that in the control group＇s skin（P0.05）.Conclusion The proportion of Th17 cells in peripheral blood and lesions of patients with psoriasis vulgaris significantly increased,and the levels of IL-17 and IL-22 both went up.
