摘 要:应用微卫星技术对云南半细毛羊、凉山半细毛羊、贵州半细毛羊、彭波半细毛羊、罗姆尼羊和考力代羊基因组DNA进行了17个微卫星位点多态性分析,以期为半细毛羊品种的遗传关系做出评价,同时为这些品种的合理开发利用提供理论依据。研究结果表明:6个研究群体PIC值均大于0.5,表明遗传多样性都极为丰富;遗传分化关系的研究表明,6个群体Nm值均在1以上,Fst值均在0.6以下,说明群体间可能存在一定的基因迁移或者有较近的亲缘关系,其中贵州半细毛羊和考力代羊间存在最大的基因流(Nm=12.065)、最小的变异系数(Fst=0.020 3)、最高的遗传一致度(0.862)及最小的遗传距离(0.148),显示出两个群体较近的亲缘关系;UPGMA聚类图显示6个群体聚为两个大支,贵州半细毛羊、考力代羊、彭波半细毛羊聚为一支,云南半细毛羊、罗姆尼羊及凉山半细毛羊聚为另一支。The genetic diversity of 6 semi-fine wool sheep breeds, including Yunnan semi-fine wool sheep, Guizhou semi-fine wool sheep, Liangshan semi-fine wool sheep, Pengbo semi-fine wool sheep, Corriedale sheep and Romney sheep was studied by 17 microsatellites loci.The result showed that the PIC value of the six breeds were above 0.5, indicating very abundant genetic di- versity of these breeds. The genetic differentiation demonstrated that there were gene flows or close relationship existing in the six breeds, with Nm〉l and Fst〈0.6. The genetic differentiation also suggested that there was a closer phylogenetic relationship be- tween Corriedale sheep and Guizhou semi-fine wool sheep with the highest level of genetic identity(0.862), the lowest genetic differentiation(Fst = 0.020 3 ), and the minimum genetic distance(0.148 ) among the six breeds. The UPGMA dendrogram indi- cated that the six breeds belonged to two different groups. The first group consisted of Guizhou semi-fine wool sheep, Corriedale sheep and Pengbo semi-fine wool sheep;the second, group included Yunnnan semi-fine wool sheep, Romney sheep and Liang- shan semi-fine wool sheep. There is farther distance between the two groups.
