摘 要:目的:观察高脂血症对大鼠脑组织基质金属蛋白酶-2(MMP-2)表达及新生血管的影响。方法:20只大鼠随机分为正常对照组和高脂血症组,每组10只。用高脂饲料喂养大鼠制备高脂血症大鼠模型。用ELISA法检测大鼠血清VEGF的含量;免疫组化法测定脑组织MMP-2、细胞间黏附分子-1(ICAM-1)、血管内皮生长因子(VEGF)和CD34的表达情况;HE染色后观察大鼠脑组织皮质区病理学改变。结果:高脂血症组大鼠血清中总胆固醇、甘油三酯和低密度脂蛋白及VEGF含量高于正常对照组(t=12.772、25.705、38.141、15.879,P均<0.001),高密度脂蛋白减低(t=35.959,P<0.001);脑组织中MMP-2、ICAM-1、CD34和VEGF的表达升高(t=25.546、12.954、8.685、11.996,P均<0.001)。结论:高脂血症可诱导脑组织表达MMP-2并生成较多的新生血管。Aim:To observe the effects of hyperlipidemia on matrix metalloproteinase-2 ( MMP-2 ) expression and the new vessels in brain tissue of rats .Methods:Experimental rats were divided into normal control group and hyperlipidemia group with 10 rats in each group .The concentration of VEGF in serum of the two groups was determined by ELISA .The ex-pressions of MMP-2, intercellular adhesion molecule-1(ICAM-1), CD34, and vascular endothelial growth factor (VEGF) in rat brain tissue were detected by immunohistochemistry .The pathological change in cortex of rats was observed after HE staining.Results:The contents of TC, TG,LDL-C and VEGF in serum of hyperlipidemia group were higher than those of normal control group(t=12.772, 25.705, 38.141, 15.879,P〈0.001), while HDL-C was lower (t =35.959,P〈0.001).The expressions of MMP-2, ICAM-1, CD34 and VEGF in brain tissue of hyperlipidemia group were higher than those of normal control group (t=25.546, 12.954, 8.685, 11.996,P〈0.001).Conclusion:Hyperlipidemia could in-duce the expression of MMP-2 and generate more new blood vessels in brain
