脂联素/肿瘤坏死因子水平对胰岛素抵抗大鼠模型脂质代谢的影响

摘  要:目的用人参皂甙Re、丹参酮IIA对高脂饮食诱导的胰岛素抵抗模型大鼠进行脂代谢干预,探讨脂联素/肿瘤坏死因子在脂质代谢中的作用。方法将SD雄性大鼠分为6组:正常组、高脂组、Re组、丹参酮IIA组、Re+丹参酮IIA组、对照组,并予以高脂饲料喂养造成胰岛素抵抗模型,喂养2 W后,开始进行药物干预,即Re组:Re 50 mg/(kg·d);丹参组:丹参酮IIA 5 mg/(kg·d);Re+丹参组:Re 50 mg/(kg·d)+丹参酮IIA 5mg/(kg·d);对照组:辛伐他汀7 mg/(kg·d),灌胃8 W。收集血液样本、肝脏、骨骼肌,脂肪组织,比较三种药物配伍对脂质代谢的作用及机制的不同。结果①各组间体重增量变化无显著性差异,Re与丹参酮IIA单独或联合使用可延缓模型鼠体重增加趋势,但其作用不如辛伐他汀。②丹参和Re联合用药可使肝脏中的脂联素受体(ADP)含量增加,对肿瘤坏死因子(TNF-α)的作用有降低趋势,单独用药可使肝脏中脂联素受体含量明显增加,对肿瘤坏死因子下降无明显作用。高脂组和中药用药组有显著性差异(P<0.05),Re+丹参组和Re组、对照组、高脂组、正常组比较均有显著性差异(P<0.05)。③在肌肉组织中脂联素受体含量以Re组较其他各组有显著性差异(P<0.05),优于对照组和高脂组,肿瘤坏死因子含量各组间无显著性差异。结论人参皂甙Re和丹参酮ⅡA联合用药对大鼠肝脏中ADP受体2含量有显著的提高作用,对大鼠肌肉中ADP受体2含量作用不及丹参酮ⅡA单用效果明显,但均优于正常组和高脂组。对TNF-α含量的调控在大鼠肝脏中联合用药能使较高的数值降低,在大鼠肌肉中较正常组降低,但各用药组之间无特异性差别。Objective to discuss the role of adiponectin and tumor necrosis factor in the metabolism of lipids from the lipid metabolism in rat model of intervention by ginsenoside Re, tanshinone ⅡA on insulin resistance induced by high - fat diet. Blood samples were collected, liver, skeletal muscle, adipose tissue, and the mechanism of three kinds of drugs on lipid metabolism and the role of different. Results there was no significant difference in weight gain betweenRe and changes of each group, tanshinone ⅡA alone or in combination can delay model rats weight increased, but its role as simvastatin. 2) Danshen and Re combination can make the adiponectin receptors in liver (ADP) content increased, on tumor necrosis factor (TNF- α) in the role of decreasing trend, alone can make the adiponectin receptor content in the liver increased significantly, decreased had no obvious effect on tumor necrosis factor. High fat group and Chinese medicine treatment group had significant difference ( P 〈 0.05), Re + Danshen group and Re group, control group, high fat group, the normal group were significantly different ( P 〈 0.05). (3) adiponectin in muscle tissue receptor content in the Re group than in other groups had significant difference ( P 〈 0.05 ), better than that of the control group and high fat group, there was no significant difference between the groups of content of tumor necrosis factor.
