摘 要:目的:观察青稞麦绿素(HBG)对小鼠免疫性肝损伤的保护作用.方法:先用HBG预处理,再采用卡介苗和脂多糖(BCG+LPS)制作小鼠急性免疫性肝损伤模型,以联苯双酯滴丸(BDD)为阳性对照.采用全自动生化分析仪测定血清谷丙转氨酶(ALT)、血清碱性磷酸酶(ALP)、血清谷草转氨酶(AST)和血清乳酸脱氢酶(LDH)的含量;连苯三酚自氧化法测定血清超氧化物歧化酶(SOD)活性;硫代巴比妥酸(TBA)显色法测定肝匀浆中的丙二醛(MDA)含量;肝组织常规制片,组织学观察.结果:不同剂量的HBG组血清ALT、ALP、AST、LDH和肝组织中MDA含量明显低于对照组,血清SOD明显高于对照组;镜下病理显示,HBG对由BCG+LPS引起的小鼠急性肝损伤造成的肝细胞水样变性、大片坏死、炎性细胞浸润等状态有缓解作用.结论:HBG对由BCG+LPS引起的小鼠免疫性肝损伤有保护作用,其作用机制可能与其抗自由基和参与免疫调节有关.Objective :To study the protective effects and the mechanism of Highland Barley Green on a- cute immune liver injury caused by BCG+LPS in mice. Methods:The mice were pretreated by Highland Barley Green (HBG). The acute immune liver injury model was mode by BCG+ LPS from tail-vein of mice. Biphenyldimethyl Diearboxylate (BDD) was as the positive control. The blood and livers were collected. The enzyme contents of ALT, ALP, AST and LDH in serum were measured by automatic biochemistry analyzer, and the activity of SOD in serum was measured by self-oxidation of pyrogallie acid, and the con- tent of MDA in liver homogenate was measured by TBA colorimetric assay. The pathological changes in liver tissue were observed. Result:Compared with group of BCG+ LPS. The contents of ALT, ALP, AST and LDH in serum and the content of MDA in liver tissue of the HBG groups were significantly lower than the control group, but also the activity of SOD was significantly higher in serum. The pathological section shows that the HBG groups protect the injury such as hydropic degeneration of liver cells, large areas of necrosis and inflammatory cell infiltration which the acute liver injury in mice was caused by BCG+ LPS. Conclusion:The HBG can protect the acute liver injury by BCG+LPS in Mice. The mechanism may be re- lated to anti-free radical and immune regulation.
