摘  要:脱除蛋白质是真菌多糖纯化过程中较为重要的环节之一,为了优化裂盖马鞍菌多糖脱蛋白质工艺,采用Sevag法和三氯乙酸法对其多糖进行蛋白质脱除,通过比较2种方法处理后的蛋白质脱除率和多糖损失率来确定最佳的脱蛋白质方法。结果表明,三氯乙酸法脱除蛋白质效果最佳,其最佳工艺为三氯乙酸质量浓度50g/L、三氯乙酸与粗多糖体积比1∶1、振荡时间40min、静置时间40min,在此条件下蛋白质脱除率为92.37%,多糖损失率为16.91%。Deproteinization is one of the most important steps in the purification process of fungi polysaccharide. In order to improve purificatiori efficiency of polysaccharide, the optimization of deproteinization process of crude polysaccharide from Helvella leucopus Pers. was carried out using Scrag reagent and trichloroacetie acid with protein removal rate and sugar loss rate as in- dexs. The results showed that trichloroacetic acid was more suitable to remove the protein from the crude polysaccharide,and the optimum trichloroacetic acid concentration was 50 g/L, the vo- lume ratio of trichloroacetic acid to crude polysaccharide was 1 : 1, vibrating time and standing time were both 40 rain. Under this condition,th'e protein removal rate was 92.37% ,and sugar loss rate was 16.91%.
