摘 要:目的探讨染砷大鼠脑组织中砷代谢产物与砷代谢相关酶的性别差异。方法将70只健康7周龄清洁级Wistar大鼠随机分为7组,分别为对照(去离子水)组及20、6.7、2.2 mg/kg亚砷酸钠和砷酸钠染毒组,每组10只,雌雄各半。采用自由饮水方式进行染毒,连续染毒3个月。测定大鼠脑组织中iAs3+、iAs5+、一甲基胂酸(MMA)、二甲基胂酸(DMA)含量及砷代谢相关酶甲基转移酶(As3mt)活力和谷胱甘肽(GSH)含量。结果各染毒组均未检出iAs5+。与对照组比较,除雌性20 mg/kg亚砷酸钠和砷酸钠染毒组大鼠脑组织中iAs3+含量低于对照组外,雌、雄其他各染毒组iAs3+含量均高于对照组(P<0.05)。与雌性大鼠比较,雄性大鼠20 mg/kg亚砷酸钠和砷酸钠染毒组脑组织中iAs3+含量较高,其他各染毒组雄性iAs3+含量均较低(P<0.05)。与对照组比较,除雌性2.2 mg/kg砷酸钠染毒组及雄性2.2 mg/kg亚砷酸钠、6.7 mg/kg砷酸钠染毒组大鼠脑组织中DMA含量未检出外,其他雌雄各剂量组有机砷含量均高于对照组(P<0.05)。与雌性大鼠比较,雄性2.2、20 mg/kg亚砷酸钠染毒组和6.7、20 mg/kg砷酸钠染毒组大鼠脑组织中DMA含量较低,雄性亚砷酸钠2.2 mg/kg及砷酸钠6.7、20 mg/kg染毒组大鼠脑组织中MMA含量较高(P<0.05)。与对照组比较,雌性2.2 mg/kg亚砷酸钠、各剂量砷酸钠染毒组以及雄性各剂量染毒组大鼠脑组织中As3mt活力均较高,雄性2.2 mg/kg、6.7 mg/kg亚砷酸钠染毒组和2.2 mg/kg砷酸钠染毒组以及雌性各剂量染毒组大鼠脑组织中GSH含量均较低(P<0.05)。与雌性大鼠比较,雄性仅2.2 mg/kg亚砷酸钠染毒组大鼠脑组织中As3mt活力和雄性6.7 mg/kg亚砷酸钠染毒组大鼠脑组织中GSH含量较低(P<0.05)。结论大鼠脑砷代谢过程中代谢产物及代谢酶雌雄间有明显差异,可能与大鼠脑砷代谢基础及脑砷甲基化能力雌雄间差异有关。Objective To explore the gender differences in arsenic metabolites of arsenic and arsenic metabolism in rat brain related enzymes. Methods A total of 70 Wistar rats aged seven weeks were randomly divided into seven groups, including control (deionized water) group and 20,6.7 and 2.2 mg/kg body weight sodium arsenite and arsenate exposure group, 10 rats in each group,half male and half female. The reatment was conducted through drinking water for consecutive three months, iAs3+, iAs5+, monomethylarsenic acid (MMA two), monomethylarsonic acid (DMA) and arsenic metabolism related enzyme methyhransferase (As3mt) activity and glutathione (GSH) content in the brain in rats were determined. Results iAs5+ was not detected in all groups. Compared with the control group, except for lower iAs3~ group, the contents of iAs3~ in the other groups were significantly higher (P〈0.05). Compared with female rats,in 20 mg/kg group, the content of iAs3+ was higher in male rats, in the other groups, they were lower (P〈0.05). Compared with the control group,organic arsenic content was higher in some exposed groups (P〈0.05). Compared with female rats,the changes of DMA and MMA in male rats in different groups were different. Compared with the control group, the changes of activity of As3mt and GSH were different in different exposed groups and gender groups. Conclusion In the rat brain, arsenic metabolites and metabolic enzyme activity are obviously different between male and female,which may be related to the differences in arsenic metabolism and cerebral arsenic methylation capacity between male and female.
