丹墨胶囊对正常大鼠组织11β-羟基类固醇脱氢酶及己糖-6-磷酸脱氢酶基因表达的影响

摘　　要：目的考察丹墨胶囊(墨旱莲、丹参、女贞子、益母草、金银花、茜草等)对大鼠11β-羟基类固醇脱氢酶(11β-HSD)和己糖-6-磷酸脱氢酶(H6PD)基因表达的影响。方法 12只雄性SD大鼠连续14 d灌胃给予丹墨胶囊0.432 g/kg后,定量RT-PCR法测定大鼠肝、肾、胸腺、脾、脂肪、肌肉、心脏、肺、小肠、睾丸等组织11β-HSD1、11β-HSD2、H6PD基因表达。结果丹墨胶囊显著诱导肝脏组织11β-HSD1基因表达,抑制大鼠肾脏、胸腺、脾、脂肪、肌肉、小肠、心脏组织11β-HSD1的基因表达。显著诱导大鼠肾脏、小肠、睾丸组织11β-HSD2的基因表达,抑制肝脏、心脏、脾、肌肉组织11β-HSD2基因表达。显著诱导肌肉、睾丸组织H6PD基因表达,抑制肝脏、肾脏、肺、脾、胸腺组织H6PD基因表达。结论丹墨胶囊可通过影响大鼠组织中11β-HSD1、11β-HSD2及H6PD基因表达,影响糖皮质激素体内活化及局部组织中激素水平。AIM To investigate the effect of Danmo Capsules （Ecliptae Herba, Salviae miltiorrhizae Radix et Rhizoma, Ligustri lucidi Fructus, Leonuri Herba, Lonicerae japonicae Flos, Rubiae Radix et Rhizoma, etc. ） on gene expression of 11 β-hydroxysteroid dehydrogenase （ 11 β-HSD） and hexose 6-phosphate dehydrogenase （H6PD） in rats. METHODS After oral administration of Danmo Capsules （0. 432 g/kg） to 12 male SD rats for 14 con- secutive days, gene expressions of 11β-HSD1, 11β-HSD2 and H6PD in tissues of liver, kidney, thymus, spleen, fat, muscle, heart, lungs, small intestine and testis were determined with quantitative RT-PCR method. RE- SULTS Danmo Capsules significantly induced gene expression of 11 ^-HSD1 in liver tissues, and inhibited gene expression of 11β-HSD1 in kidney, thymus, spleen, fat, muscle, small intestine and heart tissues. Danmo Cap- sules significantly induced gene expressions of 11β-HSD2 in kidney, small intestine and testis tissues, and inhibi- ted gene expression of 11 β-HSD2 in liver, heart, spleen and muscles. Danmo Capsules significantly induced gene expressions of H6PD in muscle and testis tissues, and inhibited gene expressions of H6PD in liver, kidney, lung, spleen and thymus tissues. CONCLUSION Danmo Capsules could change the activity of corresponding enzyme through affecting gene expression of 11β-HSD1, 11β-HSD2 and H6PD in rat tissues, and affect in vivo activation of glucocorticoids and hormone levels in local tissues.
