摘  要:目的研究神清滴丸(β-细辛醚和丁香酚)的遗传毒性。方法采用细菌回复突变试验(Ames)、中国仓鼠肺细胞染色体畸变试验(CAvit)和小鼠骨髓细胞微核试验(MNT)对神清滴丸进行安全性评估。结果 Ames试验中,在±S9mix条件下,神清滴丸各剂量组对细菌回复突变菌落数与溶媒对照组比较无统计学意义(P>0.05);CAvit试验中,在-S9mix条件下,高剂量组(24 h)和各给药剂量组(48 h)细胞畸变率均高于溶媒对照组,存在统计学意义(P<0.05);在+S9mix条件下,各剂量组(24 h与48 h)细胞畸变率与溶媒对照组比较无统计学意义。MNT试验中,神清滴丸各剂量组嗜多染红细胞比例及微核的嗜多染红细胞细胞比率与溶媒对照组比较均无统计学意义(P>0.05)。结论在本实验条件下,神清滴丸对伤寒杆菌株均无诱发致基因突变作用;在-S9min条件下,CAvit试验结果为疑似阳性,而在+S9mix条件下,结果为阴性;MNT评估亦为阴性。AIM To explore the genotoxicity of Shenqing Dripping Pills ( β-asarone and eugenol). METH- ODS Safety evaluations of Shenqing Dripping Pills consisted of the bacterial reverse mutation test (Ames test) , the chromosome aberration test (CAvit test) and the micronucleus test (MNT). RESLUTS There were no statis- tically significant increases in the number of reverant colonies at different test concentrations of Shenqing Dripping Pills with and without S9mix ( P 〉 0. 05 ). In the CAvit test, cell aberration rate from 24 h treatment with the high- est concentration of Shenqing Dripping Pills and 48 h treatment with concentrations of Shenqing Dripping Pills was higher than that from the solvent control groups without S9mix (P 〈 0. 05 ). However, there was no statistical sig- nificance in both groups with S9mix (P 〉 0. 05 ). In MNT, these was no significance in polychromatic erythrocytes count and their aberration rate between Shenqing Dripping Pills and solvent control groups ( P 〉 0. 05 ). CON- CLUSION No positive mutagenic response is observed in gene mutation induced by Salmonella typhimurium of Shenqing Dripping Pills. The CAvit test is probable positive without S9mix, whereas it is negative with S9mix and the micronuclenus test is negative.
